The expression profile and localization of genes for cytokinin-dependent cytochrome P450 monooxygenase in barley.

Abstract:

Cytokininy (CKs) jsou nízkomolekulární látky ze skupiny fytohormonů, které mají klíčovou roli v regulaci různých procesů růstu a vývoje rostlin, např. buněčného dělení, senescence listů, apikální dominance a klíčení. Z chemického hlediska jsou CKs deriváty adeninu, které mají isoprenoidní nebo aromatický postranní řetězec na N6 konci. Isoprenoidní cytokinin, trans-zeatin, má zásadní fyziologickou funkci v rostlině. Hlavním enzymem katalyzující vznik trans-zeatinu je cytokinin-dependentní cytochrom P450 monooxygenasa (CYP450). Bylo zjištěno, že nedostatek aktivity tohoto enzymu v Arabidopsis thaliana způsobuje zpomalení růstu nadzemní části rostliny. V této práci byla popsána exprese dvou genů kódujících ječmenný cytokinin-dependentní CYP450, ortologních k těmto genům v A. thaliana, v různých vývojových stádiích ječmene. Nejvyšší exprese těchto genů byla naměřena v kořenech a klasech. Dále byla sledována subcelulární lokalizace ječmenných a kukuřičných cytokinin-dependentních CYP450 - GFP fúzních proteinů v transgenních listech a transgenních ječmenných a kukuřičných protoplastech. Ve všech případech byly sledované fúzní proteiny lokalizovány v endoplazmatickém retikulu.

Abstract:

Cytokinins (CKs) are low-molecular derivatives from the group of phytohormones, which play a key role in the regulation of various processes of plant growth and development, for example cell division, leaf senescence, apical dominance and germination. From chemical point of view, CKs are derivates of adenine with isoprenoid or aromatic side chain at the N6 terminus. Trans-zeatin is an isoprenoid CK that has an essential physiological function in the plant. The main enzyme that catalyze the production of trans-zeatin is cytokinin-dependent cytochrome P450 monooxygenase (CYP450). Deficiency of CYP450 activity lead to the decrease of shoot growth in Arabidopsis thaliana. In present work, the expression of two barley cytokinin-dependent CYP450, orthologue to the A. thaliana CYP450 genes, in different developmental stages of barley was described. The highest expression of both genes was measured in roots and mature grains. In addition, subcellular localization of barley and maize CYP450 - GFP fusion proteins was performed in transgenic barley leaves and transgenic maize and barley protoplasts. The three analyzed proteins were localized in the endoplasmic reticulum in all cases.
