Mitochondria-dependent Cell Death

Abstract:

Tato bakalářská práce popisuje mitochondrie, jejich vliv na buněčnou smrt a jejich zánik. V úvodní části jsou stručně popsány základní informace a evoluční vývoj mitochondrií, dále jejich struktura, transport proteinu přes mitochondriální membrány, dynamika, a především funkce mitochondrií se zaměřením na syntézu ATP. V druhé kapitole je popsána buněčná smrt, apoptóza a nekróza. V první části této podkapitoly jsou základními informacemi o apoptóze, nejdůležitější proteiny do ní zapojené, kaspázy a Bcl-2, a dále byly popsány obě cesty apoptózy. V třetí kapitole je popsána mitoptóza tedy program smrti postihující mitochondrie. V první části je stručný popis mitoptózy vnitřní a vnější mitochondriální membrány. V druhé časti je vysvětlen mechanismus uvolňování mitochondrií z buňky a mitoptóza spojená s likvidací mitochondrií uvnitř buňky. V poslední kapitole jsou popsána mitochondriální onemocnění způsobená mutacemi v mitochondriální DNA.

Abstract:

This bachelor thesis describes mitochondria, their influence on cell death and their demise. In the introductory part is the basic information and evolutionary development of mitochondria are briefly described, as well as their structure, protein transport across mitochondrial membranes, dynamics, and especially mitochondrial function with a focus on ATP synthesis. The second chapter describes cell death, apoptosis and necrosis. In the first part of this subchapter are described basic information about apoptosis, the most important proteins involved in it, caspases and Bcl-2, and the two pathways of apoptosis. In the third chapter is described mitoptosis, a death program affecting mitochondria. In the first part is given a brief description of mitoptosis of the inner and outer mitochondrial membrane. The second part explains the mechanism of mitochondrial release from the cell and the mitoptosis associated with the destruction of mitochondria inside the cell. In the last chapter are described mitochondrial diseases caused by mutations in mitochondrial DNA.
