Analysis of essential oils in herbal beverages using single-drop microextraction technique

Abstract:

Tato diplomová práce se zabývá analýzou silic v bylinných nápojích s využitím mikroextrakce jednou kapkou metodou přímého vzorkování. Výsledky byly porovnány s analýzou byliny, při které byla použita mikroextrakce jednou kapkou metodou headspace. Získané složky silic byly analyzovány na plynovém chromatografu s plamenovým ionizačním detektorem. Analyzováno bylo deset vzorků bylinných čajů. Teoretická část popisuje vliv silic na zdraví člověka a využití bylin v běžném životě. Dále je popsán čaj jako nápoj, jeho historie, složení a výroba. V neposlední řadě jsou popsány různé extrakční a mikroextrakční techniky. Důraz je kladen na metodu mikroextrakce kapalnou fází, její historii, princip, instrumentaci a aplikace v různých odvětvích. Experimentální část je zaměřena na použité pracovní postupy, přístroje a chemikálie. Výsledky a diskuze se zabývají optimalizací teplotního programu a podmínek mikroextrakce jednou kapkou. Kvantitativní zastoupení silic v bylinných nápojích bylo vyhodnoceno metodou kalibrační křivky. Kvalitativní zastoupení silic v bylinných nálevech i bylinách bylo vyhodnoceno metodou standardního přídavku.

Abstract:

This thesis deals with the essential oils analysis in herbal beverages using direct immersion single-drop microextraction method (DI-SDME). The obtained results were compared to those obtained by headspace single-drop microextraction method from herbs. Gained essential oils were analysed by gas chromatography (GC) with flame-ionization detector. There were analyzed ten herbal tea samples. The theoretical part describes the influence of essential oils on human health and their use in daily life. There is also described tea as famous beverage, history of tea, its chemical composition and process of tea production. Finally, there are described various extraction and microextraction techniques. The special interest is dedicated to the liquid phase microextraction methods, their history, basic principles, instrumentation and application to the various kinds of samples. Experimental part is focused on the description of individual working processes, equipments and chemicals. Results and discussion part deals with the optimization of temperature program for GC as well as conditions of DI-SDME. The quantitative compositions of essential oils in herbal infusions were evaluated by calibration curve method. The qualitative compositions of essential oils were evaluated using the standard addition method.
