Molecular genetics of frontotemporal dementia and Alzheimer´s disease

Abstract:

V mé bakalářské práci se zabývám genetickou podstatou dvou typů demencí ? frontotemporální demence a demence u Alzheimerovy choroby. V rámci praktické části práce jsem vyšetřila 200 kontrolních vzorků DNA ke zjištění frekvence jednonukleotidového polymorfismu c.264+21G>A v intronu 3  genu GRN. Patogenní mutace v tomto genu jsou jednou z hlavních příčin familiární formy frontotemporální demence a současné studie uvádějí i případy Alzheimerovy choroby s nálezem těchto mutací. V následujícím textu se detailněji zabývám neuropatologickými změnami a klinickými příznaky daných onemocnění.

Abstract:

In my bachelor thesis I deal with the genetic base of two types of dementias - frontotemporal dementia and Alzheimer's disease dementia. Thus, in the practical part I evaluated 200 DNA control samples to determine the frequency of a single nucleotide polymorphism c.264+21G>A in the intron 3 of the GRN gene. Pathogenic mutations in this gene are a major cause of familial frontotemporal dementia and current studies also report the presence of mutations in this gene in Alzheimer?s disease cases. In the following text in deal with neuropatological changes and clinical signs of this diseases in more detail.
