Characterization of molecular changes coupled to differentiation pathway in human embryonic stem cells

Abstract:

Disertační práce se zabývá studiem molekulárních mechanismů charakterizujících vstup lidských embryonálních kmenových (EK) buněk do diferenciační dráhy. Detailní charakteristika molekulárních změn v řízení buněčného cyklu v průběhu diferenciace EK buněk do neurálních progenitorů je doplněna o analýzu expresního profilu microRNA. Dále práce poukazuje na souvislost signální dráhy Notch s regulací buněčného cyklu a diferenciace. Ovlivněním této dráhy je možno významně změnit čas potřebný na terminální diferenciaci neurálních prekurzorů. V závěrečné části práce poukazuje na důležitou roli microRNA nejen v řízení buněčného cyklu ale také v řízení buněčné odpovědi na poškození DNA.

Abstract:

This thesis provides an insight into molecular mechanisms that accompany process of differentiation of hESCs into specific cell type. We show detailed description of changes in cell cycle regulatory molecules that accompany process of differentiation of human ES cells into neural precursor cells (NPCs). Our study points to an interesting molecular phenomenon which might be an essential parameter of prolonged proliferation of NPCs derived from human ES cells. Additionally, we also identified several microRNAs which are coupled to induction of differentiation of hESC into NPCs, but their role in regulation of differentiation and/or cell cycle progression will have to be further investigated. Furthermore, we demonstrate that cell cycle progression is directly connected to Notch signaling. Interestingly, this finding provides an efficient tool for controlling neuronal differentiation both in vitro and in vivo. Most importantly, data also show that microRNAs play a key role in regulation of G1/S transition under self-renewing as well as DNA-damaging conditions. For the first time we provide an explanation why p21 protein is undetectable in undifferentiated human ES cells despite the presence of p21 mRNA. Moreover, we unravel a molecular mechanism that prevents p53-p21 axis of G1/S checkpoint pathway to be fully executed upon DNA damage induction.
