Effect of cyanobacterial metabolites on neuronal differentiation

Abstract:

Vyšší frekvence výskytu a přemnožení sinic je rozšířeným problémem jak ve světě, tak v České republice. Sinicové metabolity jsou bioaktivní látky, které mají potenciál ovlivňovat zdraví organismů. Pro hodnocení jejich vlivu na nejranější stádium neurálního vývoje byly vybrány dva retinoidy a sinicový toxin anatoxin-a. Těmto látkám byly exponovány lidské neurální kmenové buňky (Neural Stem Cells - NSCs) diferencující do progenitorových buněk, neuronů, astrocytů, oligodendrocytů a neuronálních subtypů. V získaných vzorcích byla analyzována exprese genů a proteinů charakteristických pro zmíněné buněčné typy (4., 13. a 22. den in vitro diferenciace) a sledována morfologie formující se neurální sítě. Bylo zjištěno, že retinoidy by mohly mít vliv na rovnováhu mezi proliferací a diferenciací neurálních buněk již od prvních dnů embryonálního vývoje. Efekt expozice neurotoxinu ATX-a nepřevažoval vliv atRA, který byl ve většině případů významnější.

Abstract:

Higher frequency of cyanobacteria occurrence and their overgrowth is a widespread problem both in the world and in the Czech Republic. Secondary metabolites produced by cyanobacteria are bioactive substances that have the potential to affect the health of organisms. Two retinoids and the cyanobacterial toxin anatoxin-a were selected to test the effect of metabolites on the earliest stage of neural development. Human Neural Stem Cells (NSCs) differentiating into progenitor cells, neurons, astrocytes, oligodendrocytes and neuronal subtypes were exposed to these substances. Subsequently, the expression of genes and proteins characteristic of neural cell types (after 4, 13 and 22 days of in vitro differentiation) was analysed and the morphology of the forming neural network was monitored it was found that retinoic acids could affect the balance between neural cell proliferation and differentiation from the very beginning of embryonic development. The effect of ATX-α exposure did not outweigh the effect of atRA, which was usually more significant.
