Prolyl-hydroxyproline Dipeptide in Non-hydrolyzed Urine - its Determination and Possible Role in Osteopathies

Abstract:

Problematika. Předložená práce se tematicky zaměřila na stanovení močového dipeptidu L-prolyl-L-4-hydroxyprolin v ranní nehydrolyzované moči plynovou chromatografií. Tato sloučenina se prokazuje být perspektivním osteomarkerem, její imunochemické stanovení však není k dispozici. Metodika. Pilotní studie použila k analýze techniku kapalinové chromatografie po přímé derivatizaci látky v moči. Pro stanovení chromatografií plynovou byla látka podrobena reakci s chlormravenčany, močové aminokyseliny a dipeptid však byly z moče předem extrahovány na pevné fázi. Záměrem této studie je prověřit možnost stanovení dipeptidu v moči přímo, bez předchozí izolace látek z matrice. K tomuto účelu byly testovány čtyři různé chlorformiáty, k analýze produktů byla použita kapilární plynová chromatografie s plamenoionizační detekcí. Dokonalé oddělení analytu a přidaného interního standardu (5-fluortryptofan) od jiných složek matrice vyžadovalo testování různých kapilárních kolon. Výsledky. Statisticky srovnatelné výsledky se stanovením kapalinovou chromatografií poskytl postup používající k derivatizaci močové matrice trifluorethylchlorformiát; korelace mezi oběma postupy (vážená Demingova regrese) poskytla hodnotu r = 0,984. Závěr. Příprava vzorku pro stanovení oběma chromatografickými technikami je rychlá a nenáročná a probíhá přímo v moči (20-50 ul), samotná analýza je v obou případech ukončena za dobu <10 min. Pracoviště však musí disponovat plynovým nebo kapalinovým chromatografem, ve druhém případě s fluorimetrickým detektorem.

Abstract:

Background. The diploma work focused on determination of urinary dipeptide L-prolyl-L-4-hydroxyproline in the morning non-hydrolyzed urine by means of gas chromatography. This compound proved to be a novel promising osteomarker, however, immuno-analytical assay of the compound is not available. Methods. High-performance liquid chromatography after derivatizing the compound directly in the urine was used in the pilot study. For gas chromatography, a direct urine treatment with a chloroformate reagent was chosen; previously, the dipeptide and amino acids were solid-phase extracted from the urine in prior. The aim of this study was to obviate the amino acid pre-isolation, and to treat the analyte directly in the urine matrix. Four reagents were tested to this purpose, the derivatization products were analyzed by gas chromatography with flame ionization detection, for which a good separation of the product and that of the internal standard (5-fluorotryptophan) from other derivatized compounds of the urine matrix on a suitable capillary column was required. Results. Close correlation between values obtained by liquid chromatography and the gas one afforded derivatization of the urine matrix with trifluoroethyl chloroformate; the corresponding correlation factor was 0.984 (weighting Deming regression). Conclusions. Direct derivatization of urine samples (20-50 ?l) for screening the dipeptide by each of the chromatographic techniques is smooth and rapid, the process and the parent analysis take few minutes only. A liquid chromatograph with fluorescence detector and gas chromatograph with flame-ionization or mass spectrometric detection are a prerequisite.
