Influence of protecting spray on vine defense reaction

Abstract:

Postřik přípravkem obsahujícím bauxit se v dnešní době používá jako alternativní ekologická metoda ochrany révy vinné, protože podle dosavadních pokusů významně snižuje napadení révy plísní révovou. Není ovšem dobře znám mechanismus, jakým tento postřik vyvolává rezistenci révy vinné vůči tomuto patogenu. Cílem práce bylo určit vliv aplikace přípravku na obrannou reakci rostlin révy vinné. Réva vinná během obranné reakce obvykle syntetizuje nízkomolekulární antimikrobiální látku resveratrol, která patří mezi fytoalexiny. V rámci diplomové práce byla optimalizována metoda pro extrakci a stanovení obsahu resveratrolu v listech révy. Dále byl testován vliv přípravku na indukci produkce resveratrolu, kdy se nepodařilo prokázat souvislost mezi ošetřením rostlin a zvýšeným obsahem resveratrolu v listech. Během obranné reakce dochází také ke zvýšení exprese tzv. obranných genů. Obranné geny kódují proteiny, které se podílejí na aktivní obraně vůči patogenům. Byly sledovány rozdíly v expresi obranných genů PR1a, chit b a chit 4c, které kódují proteiny PR1a, bazickou a kyselou chitinasu, v listech révy po ošetření. Během infekčního tlaku, kdy výrazně vzrostla exprese těchto obranných genů v kontrolních rostlinách, vzrostla exprese genů kódujících PR1a a bazickou chitinasu v ošetřených rostlinách výrazně méně. Za podmínek konstantní nebo snížené exprese studovaných genů v kontrolních rostlinách vzrostla exprese genů PR1a a chit b v rostlinách ošetřených přípravkem obsahujícím bauxit přibližně dvakrát, exprese genu chit 4c zůstala přibližně stejná.

Abstract:

Treating with bauxite is used as an alternative ecological method of grapevine protection, because this spray results according to experiments in a significant reduction of downy mildew infection in grapevine. However, the mechanism of inducing grapevine resistance against this pathogen is not yet known. The aim of this study was to determine the effects of bauxite treatment on the defence reaction of grapevine. Within the defence reaction grapevine plants usually synthesize low molecular antimicrobial compound resveratrol. During this study the method for extraction and determination of resveratrol content in grapevine leaves was optimised. Also the effect of bauxite on resveratrol production was studied but no connection between the bauxite treatment and resveratrol content was found. Also, within the defence reaction the defence genes expression is enhanced. These defence genes encode proteins, which play role in an active defence against pathogens. The differences in gene expression of PR1a, chit b and chit 4c genes, in grapevine leaves after the treatment with bauxite were observed. These genes encode PR1a protein, basic and acidic chitinase. During the period of infection stress, when the expression of defence genes in check plants increased dramatically, the gene expression of PR1a and chit b in treated plants increased significantly less. Under the conditions of constant or lowered expression of defence genes in check plants increased the expression of the genes PR1a and chit b in treated plants approximately twice, the expression of the gene chit 4c did not change.
