Functional analysis of flowering genes in cereals

Abstract:

Reprodukční úspěch rostlin závisí na správném načasování přechodu z vegetativní do reproduktivní fáze. Proto musely rostliny vyvinout sofistikované mechanismy kontrolující kvetení za pomocí genetických drah integrujících rozlišné endogenní i environmentální signály jako jsou hormony, stáří, délka dne a teplota. Genetické dráhy jsou dobře definované u modelového organismu huseníčku rolního (Arabidopsis thaliana). Nicméně, informace o molekulárním mechanismu regulace kvetení u obilovin je limitovaná. Tato disertační práce se věnuje vernalizační dráze v agronomicky významné plodině Triticum aestivum L. a jejím předkovi T. turgidum ssp. dicoccoides Körn. Vernalizace indukuje kvetení po dostatečně dlouhé periodě nízkých teplot a zajišťuje tak, že rostliny nevykvetou před zimou. Vernalizační dráhy u pšenice se účastní tři hlavní klíčové geny: VERNALIZATION1 (VRN1), VRN2 a VRN3. Alelická variace VRN1 lokusu je asociována s významnými rozdíly v době kvetení. Na základě alelické varianty se pšenice dělí na ozimý a jarní růstový typ. Ozimá pšenice nese intaktní vrn1 alely a k urychlení kvetení vyžaduje vernalizaci. Naopak jarní pšenice disponuje alespoň jednou mutantní Vrn1 alelou s dostatečnou bazální expresí pro indukci kvetení bez působení chladu. Naše výsledky poukazují na několik úrovní regulace genu VRN1, zahrnující variabilitu v počtu kopií, zpětnou regulaci exprese VRN1 genu VRN1 proteinem a sekvenční variabilitu v oblasti promotoru a prvního intronu, narušující možná vazebná místa pro regulační elementy. S cílem studovat roli kandidátních represorů VRN1 genu byly identifikovány a charakterizovány podjednotky PRC1 a PRC2 komplexů. Připravili jsme CRISPR/Cas9 editované linie pšenice seté pro studium regulace VRN1. Nově získané poznatky vyplývající z této práce mohou být užitečné při šlechtění nových kultivarů lépe adaptovaných na konkrétní environmentální podmínky, například pomocí introgrese alel genů ovlivňujících kvetení z divokého předka, pšenice plané dvouzrnky, disponující bohatou genetickou variabilitou.

Abstract:

To ensure reproductive success, plants must precisely time the transition from a vegetative to a reproductive state. Therefore, they evolved widely interconnected genetic pathways promoting flowering in response to various endogenous and environmental cues such as hormones, age, day length, and temperature. The genetic pathways are well-defined in the model plant Arabidopsis thaliana. However, the information about the molecular mechanism of flowering regulation in cereals remains limited. This thesis addresses the vernalization pathway in agriculturally essential Triticum aestivum L. and its progenitor species, T. turgidum ssp. dicoccoides Körn. Vernalization induces flowering after a sufficient period of cold temperatures and ensures plants do not flower before winter. Three main well-described VERNALIZATION genes (VRN1 to 3) are involved in the vernalization pathway of wheat. Allelic variation at VRN1 is associated with significant flowering time differences. Based on the VRN1 alleles, wheat can be divided into winter and spring growth types. Wheat with winter growth habit carries intact vrn1 alleles and requires vernalization to accelerate the flowering. Oppositely, spring wheat possesses at least one mutant Vrn1 allele with very high basal expression inducing flowering independently on cold periods. Our results demonstrate several levels of VRN1 regulation, including copy number variation, self-regulation by altered VRN1 protein, and sequence variability in regulatory regions of promoter and the first intron disrupting the possible binding sites for putative regulatory elements. With the aim of studying the role of PRC1 and PRC2 in wheat vernalization, subunits of candidate repressor complexes PRC1 and PRC2 were characterized. We prepared CRISPR/Cas9-edited wheat lines for the VRN1 regulation study. Moreover, our findings might help improve cultivated bread wheat performance under the changing climate by introducing flowering-related alleles from the wild relative species, wild emmer wheat, harboring rich genetic variability.
