Functional analysis of ATM/p53 pathway in CLL samples and testing of their sensitivity to modern therapy

Abstract:

Diplomová práce se zabývá genetickými defekty u hematologických malignit. První část je věnována funkčnímu testování proteinu ATM pomocí sledování dráhy ATM-p53-p21 u vzorků CLL. Po paralelním ošetření buněk cytostatiky fludarabinem resp. doxorubicinem byla analyzována stabilizace hladiny proteinu p53 a také indukce exprese jeho cílového genu p21. U vzorků rozdělených do skupin podle genetických abnormalit byla hodnocena tato stabilizace a exprese a korelována s jednotlivými defekty. Zásadním výstupem práce je zavedení efektivního a spolehlivého testu pro analýzu funkčnosti proteinu ATM. Je založen na sledování indukce exprese genu p21 pomocí qRT-PCR po paralelním ošetření CLL buněk fludarabinem resp. doxorubicinem – v případě inaktivace ATM dochází k indukci po prvním, avšak nikoli po druhém cytostatiku. Ve druhé části práce byla testována senzitivizace buněčných linií SU-DHL-4 a MEC-1 s mutacemi genu TP53 k cytostatiku fludarabinu pomocí inhibice kináz ATM a Chk1. Senzitivizace byla úspěšná u obou linií při společném použití inhibitorů. U linie MEC-1 vykazovaly inhibitory částečný účinek i samostatně.

Abstract:

The major subject of this diploma thesis relates to genetic defects in hematological malignancies. The aim of the first part was a functional testing of ATM protein in CLL samples using a monitoring of the ATM-p53-p21 pathway. The samples were treated in parallel by the cytostatics fludarabine and doxorubicin. They were subsequently analyzed by western blotting and qRT-PCR. The level of stabilized p53 protein and the expression of its downstream target gene p21 were evaluated and correlated with genetic defects in studied samples. The main result of this thesis is the introduction of effective and reliable assay of ATM protein functionality. This assay is based on monitoring of p21 gene expression - if ATM is inactivated, the induction of p21 gene occurs after flurdarbine treatment, but not after doxorubicin treatment. The second aim of the thesis was a testing of potential sensitization to fludarabine using inhibition of ATM and Chk1 kinases. Cell lines SU-DHL-4 and MEC-1 harboring T53 mutation were used. Sensitization was particularly successful when using both inhibitors together. The MEC-1 cell line was partially sensitized also by individual inhibitors.
