Nucleolar dominance and expression of homeologous rDNA genes in allotetraploid Tragopogons

Abstract:

Jadérková dominance je epigenetický jev projevující se umlčením jednoho z rodičovských genomů po hybridizaci. Ke studiu tohoto jevu jsem využila nově vzniklý allotetraploidní druh, Tragopogon mirus (T. dubius x T. porrifolius). V rámci populační studie byly objeveny populace bez dominance, s kodominancí a úplnou dominancí rDNA jednotek. Variabilita mezi jedinci populací s dominantním fenotypem byla zanedbatelná. Většina populací vykazovala dominanci rDNA jednotek původem z T. dubius, které jsou v menšině, zatímco rDNA jednotky původem z druhého homeologa jsou transkripčně umlčeny. U jedné populace (2602) byl tento rys změněn na kodominantní stav. Charakter dominance může být dokonce převrácen jak jsem zjistila v kořenových špičkách rostliny 2603-33A a jiných rychle se dělících tkáních. Tato rostlina nese rozsáhlou deleci rDNA původem z T. dubius v homozygotním stavu. Domnívám se, že zatímco dominantní lokus je schopný zajistit dostatečnou transkripci rRNA v listu, není schopen pokrýt potřeby rychle se dělících buněk. Ve studovaných orgánech byla reaktivace původně umlčených jednotek doprovázena hypomethylací rDNA původem z T. porrifolius. Při použití hypomethylačbního činidla, 5-azacytidinu, byl odstraněn blok znemožňující transkripci umlčených rDNA jednotek. Lze tedy vyvodit, že transkripce rRNA genů je u allotetraploida T. mirus ovlivněna methylací DNA.

Abstract:

Interspecific hybrids and allopolyploids often silence expression of ribosomal genes inherited from one parental species. This phenomenon, known as nucleolar dominance, was studied at the population level in recent allotetraploid Tragopogon mirus (T. dubius x T. porrifolius). We have identified populations with no, partial and complete nucleolar dominance. Interindividual variability in populations with established dominant phenotype was low or negligible. Most populations showed dominance of low copy T. dubius units, while those of more abundant partner homoeologue was suppressed. However, in one population (2602) of T. mirus this general trend was reversed. I further show that dominance can be switched to codominance in root tissues (plant 2603-33A) containing metabolically highly active cells. This individual bears a homozygous macrodeletion at the transcriptionally dominant locus of T. dubius origin. I hypothesize that while the domimant locus with less copies is able to guarantee production of rRNA in leaf this amount of genes is apparently insufficient to drive rDNA expression in metabolically highly active tissues (root tip meristem,...). In these organs activation of silent genes of T. porrifolius origin was associated with hypomethylation of restriction sites within intergenic spacer. Application of a hypomethylation drug, 5-azacytidine, released transcriptional block in individuals with silent phenotype. We conclude that expression of rRNA genes is influenced by DNA methylation in Tragopogon allotetraploids.
