Honeybee as a biomonitor of silver occurrence in a locality

Abstract:

Tato diplomová práce se zabývá možností využití včely medonosné (Apis mellifera) jako biomonitoru přítomnosti stříbra (Ag) v životním prostředí. Cílem bylo ověřit, zda lze pomocí rutinního analytického postupu stanovit detekovatelný obsah stříbra v tělech včel, a využít tato data k odlišení stříbronosných lokalit od referenčních oblastí. Hypotéza práce vycházela z předpokladu, že včely medonosné, přicházejí při hledání potravy do pravidelného kontaktu s prachovými a půdními částicemi, které mohou obahovat stopy stříbra. Tyto částice mohou ulpívat na povrchu těla včel či být absorbovány do jejich tkání. V rámci laboratorního byly testovány různé metody mineralizace a složení extrakčního činidla. Jako nejvhodnější se ukázala kombinace mikrovlnné mineralizace včelích těl bez předchozí homogenizace a použití Leffortovy lučavky jako oxidačního činidla. Tato metoda byla validována na referenčním materiálu (Metranal 01). Pro vlastní stanovení obsahu Ag byla využita metoda ICP-MS, využívající reakční režim s kyslíkem (O2), což umožnilo efektivní eliminaci spektrálních interferencí, které mohou ovlivnit přesnost stanovení obsahu Ag. Výsledky ukázaly, že včely ze stříbronosných lokalit vykazovaly konzistentně vyšší obsah Ag ve srovnání s kontrolními lokalitami, kde se koncentrace pohybovaly převážně pod detekčním limitem metody (LOD). Práce tak potvrzuje možnost stanovení stopových koncentrací stříbra ve včelím těle pomocí moderních analytických technik, což může sloužit jako efektivní nástroj pro screening výskytu stříbra v prostředí. Výsledky práce mohou sloužit jako podklad pro další výzkumy i pro praxi v oblasti environmentální chemie a ochrany přírody.

Abstract:

This thesis explores the potential use of the honeybee (Apis mellifera) as a biomonitor of silver (Ag) presence in the environment. The primary objective was to determine whether a routine analytical procedure can detect trace amounts of silver in bee tissues, and whether such data can be used to distinguish Ag-enriched localities from reference areas. The working hypothesis assumed that honeybees, while foraging, come into regular contact with dust and soil particles that may contain trace levels of silver. These particles can adhere to the surface of the bees’ bodies or be partially absorbed into their tissues. In the laboratory phase, various sample digestion methods and compositions of extraction reagents were tested. The most effective approach was found to be microwave-assisted digestion of whole bee bodies (without prior homogenization), combined with the use of Leffort's aqua regia as the oxidizing agent. This method was validated using a certified reference material (Metranal 01). For the quantification of silver, inductively coupled plasma mass spectrometry (ICP-MS) was employed in oxygen (O₂) reaction mode, which enabled effective suppression of spectral interferences that could compromise the accuracy of Ag determination. The results demonstrated that bees collected from silver-bearing localities consistently exhibited higher Ag concentrations compared to those from reference areas, where concentrations were generally below the method detection limit (LOD). These findings confirm the feasibility of detecting trace levels of silver in bee tissues using advanced analytical techniques and support the use of bees as an efficient screening tool for assessing silver occurrence in the environment. The outcomes of this study may serve as a foundation for further research and practical applications in the fields of environmental chemistry and nature conservation.
