Modern approaches in neural activity analysis

Abstract:

Tato práce se věnuje metodám funkcionální analýzy, které jsou použity ke studiu neuronální aktivity krysy. Tyto metody reprezentují moderní přístup k datové analýze a umožňují zkoumat také derivace funkcí. Výsledkem pak je více získaných informací o datech. Cílem práce je vytvořit model popisující neuronální aktivitu, jejíž pochopení by mohlo pomoci vysvětlit nejasné příčiny vzniku tinitu. Neuronální aktivita je měřena pomocí potenciálů vyvolaných zvuky s různou intenzitou. Vyvolané potenciály (EP) jsou měřeny při třech různých zvukových frekvencích po dobu šesti dní a osmi kontrolních dním, kdy je u krysy vyvolán tinitus předávkováním salicátem sodným (SS). Série intenzit se skláda ze 79 sérií měření EP pro daný den a zvukovou frekvenci. Diskrétně měřená data jsou transformována do funkcionálního tvaru využitím B-splajnové báze řádu pět. Data jsou vyhlazena pomocí penalizační funkce třetího řádu, přičemž vyhlazovací parametr je zvolen pro každou sérii zvlášť pomocí kritéria zobecněné krosvalidace. Pro analýzu používáme také derivace vyhlazených funkcí. Pro každou sérii intenzit je zvolen reprezentant, kterým je medián spočítán s pomocí zobecněné pásové hloubky. Hlavní metoda analýzy neuronální aktivity je funkcionální analýza hlavních komponent. Zkoumáme v ní variabilitu mezi reprezentanty funkcí a derivací pomocí prvních dvou komponent, které v obou případech vysvětlují více než 80% celkové variability v datech. Zjisťujeme, že první komponenta odpovídá převážně rozdílům mezi zvukovými frekvencemi a druhá komponenta odpovídá hlavně rozdílům mezi kontrolní a SS léčbou. Také používáme VARIMAX kritérium pro rotaci komponent abychorstudim vylepšili jejich interpretaci. Zjisťujeme minimální rozdíly mezi komponentami před a po rotaci pro derivace funkcí. Efekty komponent pro reprezentanty původních funkcí jsou odlišné pro komponenty po rotaci, ale stále nejsme schopni rozlišit mezi vlivem léčby a vlivem zvukové frekvence. Na základě skóre hlavních komponent oddělíme kontrolní dny od SS dní a zvuk click od zbývajících dvou zvukových frekvencí. Pozorujeme slabou separaci zvukové frekvence 10kHz od 16kHz.

Abstract:

The thesis deals with the methods of the functional data analysis which are used for studying the neural activity of a rat. The functional methods represent a modern approach to the data analysis and enable examining also the derivatives of the functions. This results in obtaining more information about the data. The aim of the thesis is to create a model describing the neural activity because its understanding may help to find the unclear causes of the onset of tinnitus. The neural activity is measured as the potentials evoked by sounds of various intensities under three different sound frequencies for six control days and eight days, when the rat is overdosed with the sodium salicylate (SS) to induce the tinnitus. An intensity series consists of 79 series of EP measurements at the given day and sound frequency. The discretely measured data are transformed into the functional form using order five B-spline basis. They are smoothed using order three penalization function with the smoothing parameter chosen for each intensity series using the generalized crossvalidation. A representative is chosen for each intensity series of functions and their derivatives as the median calculated with the help of the modified band depth. The main method of the analysis of the neural activity is the functional principal component analysis. We explore the variability between the representatives of functions and derivatives using the first two components, which in both cases explain more than 80% of the total variance of the data. We find out that the first component accounts mainly for the differences between the sound frequencies and the second component for the differences between the control and SS treatment. We use the VARIMAX criterion for rotating the components to improve their interpretation. For the representatives of derivatives, there are only slight differences between the components before and after the rotation. The effects of the new components for the representatives of functions are different than before the rotation, but we are still not able to distinguish between the effect of the treatment and the effect of the sound frequency. Based on the principal components scores, we separate the control days from SS days and the click sound from other two sounds. There is a weak separation of the sound frequencies 10 kHz and 16 kHz.
