Distribution function approach to kinetic study of ligand-receptor interactions by flow cytometry and confocal laser scanning microscopy

Abstract:

Úvod V posledních letech se průtoková cytometrie a konfokální skenovací laserová mikroskopie vyvinuly na široce používané techniky zkoumání ligand-receptorových interakcí na povrchu a uvnitř buněk. Unikátní vlastností průtokové cytometrie je, že poskytuje detailní informace o nehomogenních populacích buněk ve formě cytogramů. Jednotlivé buňky jsou zobrazeny jako body v multidimenzionálním prostoru, se souřadnicemi přímého a bočního světelného rozptylu a intenzitou fluorescence v různých vlnových délkách. Navzdory této výhodě značná část potenciálu distribučně-funkčního přístupu v průtokové cytometrii zůstává nepovšimnutá, protože obyčejně se z cytogramů vyhodnocují jenom průměrné hodnoty fluorescence v populaci buněk bez ohledu na nehomogenitu populace. Konfokální skenovací laserovou mikroskopii lze s výhodou využít pro FRAP techniku (fluorescence recovery after photobleaching), která umožňuje charakterizovat dynamické chování makromolekul v různém prostředí. Nedávný pokrok ve FRAP umožňuje výzkumným pracovníkůmvypočítat relativní difuzní a vazební konstanty fluorescentně označených makromolekul v živých buňkách. Přístup však čelí problémům při analytickém rozboru měření v nehomogenním prostředí. Výsledky Byl vyvinut nový metodologický přístup pro kinetické studium ligand-receptorových interakcí v nehomogenním prostředí. Bylo potvrzeno, že kinetické údaje o ligand-receptorových interakcích získané z průtokové cytometrie mohou být interpretovány jako časový vývoj fluorescenčního profilu buňky(co se týče počtu buněk versus intenzita fluorescence), z něhož lze získat souhrnné informace o rozložení povrchových receptorů v celé buněčné populaci a též asociační a disociační vazebné konstanty jednotlivých receptorů. Byla vyvinuta nová „nonfitting“ metoda vyhodnocování průměrných efektivních difuzních koeficientů v určitých oblastechjádra buňky (která může zahrnovat celé jádro) pomocí matematické analýzy dvojdimenzionálního prostorového obrazce obnovy fluorescence. Prostorově a časově závislé výsledky měření transportu proteinů uvnitř buněk byly získány s využitím FRAP techniky. Autorka disertace svou experimentální a teoretickou prací významně přispěla k rozvoji tohoto nového metodologického přístupu. Závěry Díky kvantitativním měřením procesu ligand-receptorové interakce na nehomogenních populacích buněk mohou vypracované přístupy zlepšit naše znalosti interakcí proteinů v živých buňkách a přivedou nás tak o krok blíže k pochopení složitých proteinových sítí, které řídí chování buněk.

Abstract:

Background In recent years, flow cytometry and confocal laser scanning microscopy have emerged as widely used techniques for studying ligand-receptor interactions on the surface of and inside cells. A unique feature of flow cytometry is that it provides detailed information on inhomogeneous cell populations in the form of cytograms. Single cells are presented as points in multidimensional space, with coordinates of forward and side light scattering and fluorescence intensity at different wavelengths. In spite of this advantage, a significant part of the potential of the distribution function approach in flow cytometry is still unrealized, as usually only mean values of fluorescence over the cell population are evaluated from the cytograms for further consideration without taking into account the inhomogeneity of the population. A laser scanning confocal microscopy is a good platform for the fluorescence recovery after photobleaching (FRAP) technique, which allows the characterization of the dynamic behavior of macromolecules in different media. Recent advancements in FRAP allow researchers to quantitate diffusion and binding rate constants of fluorescently labeled macromolecules inside living cells. However, the approach poses problems in the analytical tractability of measurements in an inhomogeneous medium. Results A new methodological approach to the kinetic study of ligand-receptor interactions in inhomogeneous media was developed. It was confirmed that flow cytometric kinetic data on ligand-receptor interactions can be treated as a temporal evolution of the cellular fluorescence profile (in terms of number of cells versus fluorescence intensity) to get comprehensive information on the distribution of surface receptors in the entire cell population, as well as the association and dissociation rate constants per receptor. A new nonfitting method to evaluate the averaged effective diffusion coefficient of a region of interest in the cell nucleus (which may include a whole nucleus) by mathematical processing of the entire two-dimensional spatial pattern of recovered fluorescence was developed. Spatially and temporally resolved measurements of protein transport inside cells were obtained using the FRAP technique. The thesis author contributed substantially to the main experimental and theoretical work on the development of the above mentioned new methodological approach. Conclusions By providing quantitative measurements of the process of ligand-receptor binding in inhomogeneous cell populations, the approaches developed are believed to improve our knowledge of binding interactions between proteins in living cells, thereby bringing us a step closer to understanding the complex protein networks that regulate cellular behavior.
