Intestinal microbiome and its role in human diseases

Abstract:

Countless microorganisms, including Archaea, Bacteria, Eukarya and viruses can be found in the human body. The sum of their genomes exceeds the number of human genes in our body several times. Majority of them belongs to the intestinal microbiome, the collective complex of microorganisms colonizing the human gut. Development of the next-generation sequencing methods in recent years has revealed a close relation of the microbiome to the human health. Researches focus mainly on the connection of bacteria presence to various diseases. This bachelor thesis deals with the relationship of the intestinal microbiome to the most important illnesses, including obesity, colorectal carcinoma, non-alcoholic fatty liver diseases and inflammatory bowel diseases. It is assumed, that the key factor in this issue is the changed composition of microbiome. It often leads to the increased growth of pathogens, production of harmful substances (e.g. endotoxins, carcinogens etc.) or to increased immunostimulation. The results of experiments of the last decade open new opportunities in treatment of the diseases mentioned above.

Abstract:

V tele človeka môžeme nájsť veľké množstvo mikroorganizmov, ktoré zahŕňajú Archaea, Bacteria, Eukarya a vírusy. Súčet ich genómov niekoľkonásobne prevyšuje počet génov nášho tela. Prevažná časť z nich patrí práve intestinálnemu mikrobiómu, ktorý predstavuje kolektívny súbor mikroorganizmov osídľujúcich ľudské črevo. Rozvoj sekvenačných metód novej generácie v posledných rokoch odhalil úzky vzťah mikrobiómu a zdravia človeka. Výskumy sa zameriavajú hlavne na spojitosť výskytu baktérií a rôznych chorôb. Táto bakalárska práca sa zaoberá vzťahom intestinálneho mikrobiómu k tým najvýznamnejším ochoreniam, a to k obezite, kolorektálnemu karcinómu, nealkoholickým ochoreniam pečene a zápalovým ochoreniam čriev. Predpokladá sa, že kľúčovým faktorom v tejto problematike je zmenené zloženie mikrobiómu. To často vedie k zvýšenému rastu patogénov, produkcii škodlivých látok (napr. endotoxínov, karcinogénov a pod.), prípadne k zvýšenej imunostimulácii. Výsledky výzkumu mikrobiomu v poslednom desaťročí otvárajú nové možnosti v liečbe spomenutých ochorení.
