Study of protein phosphorylation using CELIF

Abstract:

V první části této práce bylo úkolem se seznámit s metodou CE - LIF na teoretické úrovni. V experimentální části bylo vytyčeno několik cílů. Jedním z nich bylo správně nastavit přístroj a zjistit LOD, k čemuž byl využit modelový vzorek (riboflavin). Reprodukovatelnost měření riboflavinu byla uspokojivá jak z hlediska migračního času tak výšky píku. LOD přístroje pro riboflavin byl určen na 4,1.10-9 mol.l-1. Hlavním cílem bylo ověřit a optimalizovat separační podmínky pro AHP5- GFP ve slabě alkalickém prostředí a docílit dostatečnou reprodukovatelnost výsledků. V obou případech se podařilo cíl splnit. Pro dostatečnou reprodukovatelnost výsledků bylo nezbytné kapiláru mezi jednotlivými měřeními propláchnout. Zkoumána byla i stabilita vzorku AHP5-GFP. Tento komplex je nestabilní a dochází k odfúzovávání GFP. Tuto skutečnost neovlivnil ani přídavek jednoho z inhibitoru (EDTA, PMSF). Na odfúzovávání GFP nemá vliv ani dlouhodobé skladování při -80 °C a -20 °C. Tyto výsledky byly potvrzeny western blottingem. Stabilita vzorku při uchovávání v lednici při teplotě 4 °C je dostatečná po dobu tří dnů, poté dochází ke změnám vzorku. Fosforylace se prozatím nepodařila, příčinou může být přítomnost balastních proteinů ve vzorku či nefunkčností fosforylačního postupu.

Abstract:

The first part of this thesis was getting theoretical knowledge of the CE - LIF method. Several objectives were set in the experimental part. One of them was to adjust the device properly and to determine the LOD using a model sample (riboflavin). The reproducibility of riboflavin analysis was satisfactory in the terms of both migration time and peak height. The device LOD of 4,1.10-9 mol.l-1 was determined for riboflavin. The main objective consists in the verification and optimization of the separation conditions for AHP5-GFP in a slightly alkaline environment and the achievement of the sufficient reproducibility. In both cases, the target was met successfully. For adequate reproducibility of results it was necessary to flush the capillary between the particular measurements. The stability of the sample AHP5-GFP was examined as well. The complex was unstable with GFP being released. This phenomenon was not affected by the addition of one of the inhibitors (EDTA, PMSF) either. The release of GFP was not affected even by a long-term storage at -80 ° C and -20 ° C. These results were confirmed by the western blotting. The stability of the sample during storage in a refrigerator at 4 ° C is sufficient for three days, after that there are changes of the sample.
