Optimisation of purification of human RNAP II from insect cells

Abstract:

RNA polymeráza II (RNAP II) je centrálním enzymem transkripce. Je to složitý vícepodjednotkový proteinový komplex, zodpovědný za přepis všech genů kódujících bílkoviny. Obsahuje mnoho zajímavých prvků, jako je například karboxy-terminální doména (CTD) nebo disociovatelný RPB4/7 subkomplex. Její nezpochybnitelná důležitost ji učinila předmětem výzkumu už od doby jejího objevení. Nicméně, RNAP II se liší od organismu k organismu a výzkum modelových organismů, jako Saccharomyces cerevisiae, může dopřát pouze omezené pochopení, pokud chceme porozumět struktuře a funkčním mechanismům tohoto enzymu u lidí. Pro umožnění výzkumu lidské RNAP II, je zapotřebí efektivního systému pro purifikaci tohoto proteinu. V této práci představujeme základy toho, co by se mohlo stát takovýmto systémem. Zaměřili jsme se na purifikaci disociovatelného RPB4/7 subkomplexu, RNAP II obsahující celou CTD a také RNAP II zcela postrádající CTD. Zde prezentujeme klonování a expresi lidského RPB4/7 subkomplexu v E. coli a klonování lidského RNAP II komplexu, obsahující i postrádající CTD, schopného se exprimovat v hmyzích buňkách.

Abstract:

RNA polymerase II (RNAP II) is a central enzyme of transcription. It is an intricate multisubunit protein complex, responsible for transcription of all protein coding genes. It contains many interesting features, such as the carboxy-terminal domain (CTD) or the dissociable RPB4/7 subcomplex. Its undeniable importance has made it an object of research ever since it has been discovered. However, RNAP II is known to differ from organism to organism and the research of model organisms, such as Saccharomyces cerevisiae, can provide only limited insight, if we wish to understand the structure and functional mechanisms of this enzyme in humans. To enable research of human RNAP II, an effective system for purification of this protein is required. In this thesis we present the foundation of what could become such a system. We focused on the purification of the dissociable RPB4/7 subcomplex, RNAP II containing full length CTD and also the RNAP II lacking the CTD entirely. Herein we present the cloning and expression of the human RPB4/7 subcomplex in E. coli and cloning of the human RNAP II complex, both containing and lacking the CTD, capable of expression in insect cells.
