Preparation of recombinant tetrathionate hydrolase from Acidithiobacillus ferrooxidans

Abstract:

V rámci teoretické části se tato práce zaměřuje na význam a charakteristiku extrémně acidofilních mikroorganismů, jejichž modelovým zástupcem je bakterie A. ferrooxidans. Dále se teoretická část zabývá tetrathionanhydrolasou, důležitým enzymem metabolismu anorganických sirných látek. V praktické části byla provedena optimalizace produkce rekombinantního proteinu tetrathionanhydrolasy kmene A. ferrooxidans CCM 4253 izolovaného ze Zlatých Hor. V rozporu s dosavadní literaturou se nám podařilo produkovat tento rekombinantní enzym v aktivním stavu do periplazmy E. coli. Z předběžných dat dále vyplývá, že aminokyseliny arginin a lysin nejspíše nemají vliv na aktivitu tetrathionanhydrolasy, naopak aminokyseliny histidin, tryptofan, tyrosin, serin, kyselina glutamová a asparagová se pravděpodobně podílejí na aktivitě tetrathionanhydrolasy.

Abstract:

This work focuses on the importance and characteristics of extremely acidophilic microorganisms, whose model representative is A. ferrooxidans. Furthermore, the theoretical part deals with tetrathionate hydrolase, an important enzyme involved in metabolism of reduced inorganic sulfur compounds. In the practical part, the production of the recombinant tetrathionate hydrolase of A. ferrooxidans CCM 4253 isolated from Zlaté Hory was optimized. Contrary to the literature, we have succeeded in producing the active recombinant enzyme into the periplasm of E. coli. Preliminary data also suggested that the amino acids arginine and lysine most probably have no effect on the activity of tetrathionate hydrolase, on the other hand, the amino acids histidine, tryptophan, tyrosine, serine, glutamic acid and aspartic are likely to contribute to the activity of tetrathionate hydrolase.
