EZH2 as a possible prognostic marker for colon adenocarcinomas

Abstract:

Adenokarcinom tlustého střeva patří mezi nejčastější příčiny úmrtí v rozvojových zemích. V jeho prognóze hraje důležitou roli řada prediktivních faktorů, mezi které patří staging a grading v době prvozáchytu, angioinvaze, metastazování a případně mutace v genech RAS a BRAF. V naší práci jsme se zaměřili na sledování exprese proteinu EZH2, který dle současné literatury patří mezi prognostické markery spojené s horší prognózou a progresí u různých typů maligních tumorů včetně kolorektálního karcinomu. V námi vytvořeném souboru 55 adenokarcinomů byla porovnána exprese mezi skupinou 26 metastazujících a 29 nemetastazujících karcinomů a dále mezi prognostickými skupinami stagingu (pT1-pT4) a gradingu (grade 1-3). Naše výsledky prokázaly nárůst mediánu exprese pouze mezi skupinou dobře diferencovaných (low grade) a nízce diferencovaných (high grade) karcinomů. Lze tedy soudit, že riziko progrese karcinomu se zvyšuje se stupněm diferenciace spíše než se stupněm pokročilosti tumoru.

Abstract:

Colorectal carcinoma is one of the most common causes of death in developing countries. A number of predictive factors play an important role in its prognosis, including staging and grading at the time of first capture, angioinvasion, metastasis, and possibly mutations in the RAS and BRAF genes. In our work, we focused on monitoring the expression of EZH2 protein, which according to the current literature is one of the prognostic markers associated with poorer prognosis and progression in various types of malignant tumors, including colorectal cancer. In our set of 55 adenocarcinomas, the expression was compared between a group of 26 metastatic and 29 non-metastatic carcinomas, as well as between the prognostic groups of staging (pT1-pT4) and grading (grade 1-3). Our results showed an increase in median expression only between the group of well-differentiated (low grade) and low-differentiated (high grade) tumors. Thus, it can be concluded that the risk of cancer progression increases with the degree of differentiation rather than the degree of tumor progression.
