Expression and characterization of the ferric reductase FerA from the bacterium Paracoccus denitrificans

Abstract:

FerA is a flavin reductase isolated from the cytoplasmic fraction of P. denitrificans, and most probably involved in iron assimilation. The aim of this work was to find an expression system that would allow the production of this enzyme in higher amounts and purity. The next goal was a closer characterization of this enzyme. The expression system was successfully developed, and the protein was produced in the required amount and purity with the use of C –terminal His-taging. The expression did’n work with the N-terminal His-tag. Afterwards, the kinetic constants Km and Vmax were measured for the recombinant enzyme, and they turned out to be similar compared with the native enzyme. The observed pH optimum of the enzyme was equal to the physiological value of ~7. Inhibition studies with common flavine reductase inhibitors were carried out. It was shown that cibacron blue 3GA is a potent inhibitor of rFerA, whereas dicumarol was ineffective. Thermal deactivation observations showed a rapid loss of activity above 50 oC for the recombinant enzyme. Covalent modifications of the aminoacid side chains with various reagents were investigated. The results suggested the possible presence of arginine, histidine and acidic residues in the active center.

Abstract:

FerA je flavín reduktáza izolovaná z cytoplazmovej frakcie baktérie P. denitrificans, ktorá z najväčšou pravdepodobnosťou hrá úlohu v asimilácii železa. Cieľom tejto práce bolo nájsť expresný systém ktorý by umožnil produkciu tohto enzýmu vo väčších množstvách a čistote. Druhým cieľom práce bola bližšia charakterizácia tohto enzýmu. V práci sa podarilo skompletizovať expresný systém s ktorým bol rekombinantný proteín následne produkovaný v požadovanom množstve a čistote, a to pri použití C-terminálnej His-tag kotvy. Pri N-terminálnej kotve expresia neprebiehala. Následne boli stanovené kinetické konštanty Km a Vmax pre rekombinantný enzým, pričom sa ukázalo, že sú podobné pri porovnaní s natívnym enzýmom. Bolo tiež stanovené pH optimum enzýmu, ktorého hodnota je zhodná z fyziologickým pH ~7. Následne boli vykonané testy z dvoma známymi inhibítormi flavín reduktáz, pričom cibakronová modrá 3GA sa ukázala ako účinný inhibítor rFerA. Dikumarol nebol ako inhibítor účinny. Pri sledovaní termálnej deaktivácie dochádzalo k rýchlej strate aktivity nad teplotou približne 50 oC. Boli skúšané tiež kovalentné modifikácie aminokyselinových zvyškov s viacerými činidlami, pričom namerané dáta potvrzujú alebo poukazujú na možnosť prítomnosti kyslých zvyškov, ďalej na prítomnosť arginínu a taktiež histidínu v aktívnom centre.
