Skin penetration of chemical substances

Abstract:

This thesis is concerned with the permeation experiments with skin. The aim of the thesis, to introduce a perspective and fully automatized method - Franz cell – to the Research Centre for Toxic Compounds in the Environment, has been fulfilled. The theoretical part is the review of in vitro methods for dermal absorption sudies, especially the Franz cell method and the methodology of the permeation experiments. It focuses on the description of the choice and structure of the skin used in dermal absorption studies, as well as on the importance of absorption kinetics studies for the formation and development of the models for risk assessment. Studies that approve the functionality of the automatic system for permeation experiments, as well as the assays where the absorption kinetics of two polar pesticides have been measured, have been carried out. Steady-state fluxes have been determined for chlorotoluron (34,15 ng*cm-2*h-1) and carbaryl (82,42 ng*cm-2*h-1), from which the permeability coefficients have been calculated. The results have been discussed considering the complex information about dermal absorption.

Abstract:

Práca sa zaoberá problematikou permeačných experimentov s kožou. Bol splnený cieľ zaviesť perspektívnu, plne automatizovanú metódu Franz cell do Centra pro výzkum toxických látek v prostředí. Teoretická časť práce sa zameriava na prehľad in vitro metód pre skúmanie dermálnej absorpcie, medzi nimi aj metódou Franz cell a metodikou týchto experimentov. Ďalej sa bližšie zameriava na popis výberu a štruktúry kože a význam skúmania kinetiky absorpcie pre účely vytvárania a zdokonaľovania modelov pre hodnotenie zdravotných rizík. V experimentálnej časti boli realizované štúdie potvrdzujúce funkčnosť automatického zariadenia na permeačné experimenty, a testy, v ktorých bola meraná kinetika absorpcie dvoch polárnych pesticídov. Boli zistené toky v ustálenom stave pre chlorotoluron (34,15 ng*cm-2*h-1) a carbaryl (82,42 ng*cm-2*h-1), na základe ktorých boli vypočítané ich permeačné koeficienty. Výsledky boli interpretované v širších súvislostiach problematiky dermálnej absorpcie.
