The effect of organization of pigment-protein complexes on efficiency and utilization of excitation energy

Abstract:

Tématem, kterým se zabývá tato práce, je účinnost přenosu excitační energie v závislosti na zastoupení pigment-proteinových komplexů. V teoretické části práce je rozpracována problematika struktury pigment-proteinových komplexů (PPC) v thylakoidní membráně. V dalších kapitolách jsou popsány způsoby přenosu excitační energie v PPC, které je vhodné studovat pomocí excitačních a emisních spekter. Jedna kapitola je zaměřena také na mutanty ječmene jarního, jež se používají právě pro zjišťovaní úloh jednotlivých proteinů ve složitém procesu přenosu excitační energie. Výrazný vliv na strukturu komplexů má intenzita ozářenosti a změna teplot. Poslední kapitola teoretické části se zabývá nejen vlivem uvedených stresových faktorů na jednotlivé pigment-proteinové komplexy, ale také na thylakoidní membrány a listy. V experimentální části byla nízkoteplotní fluorescencí získána emisní a excitační spektra listů divokého typu (WT) ječmene jarního (Hordeum vulgare L.) i jeho mutantu chlorina f2 aklimovaných na vysokou a nízkou ozářenost. Kromě intaktního materiálu byla také měřena spektra suspenzí thylakoidních membrán a pigment-proteinových komplexů, které byly separovány PAGE analýzou. Pomocí metody HPLC a absorpční spektrofotometrie byly stanoveny obsahy pigmentů v suspenzích thylakodních membrán i v elektroforeticky oddělených PPC. Vliv teploty za současného působení vysoké ozářenosti na účinnost přenosu excitační energie byl studován na smrku ztepilém (Picea abies L.). Byla změřena fluorescenční spektra při teplotě kapalného dusíku (77K) i při pokojové teplotě na jehlicích smrku alimovaných na různé teploty a ozářenosti. Ze suspenzí thylakoidních membrán byly použitím HPLC zjištěny obsahy pigmentů.

Abstract:

The main objective of this thesis was to study the efficiency of excitation energy transfer in dependence on structural organization of pigment-protein complexes (PPC). In theoretical part of this work is mentioned the structure of PPC in tylakoid membrane. In the next chapter the ways of excitation energy transfer, that can be studied by fluorescence spectra, are described. One section is also focused on mutants of barley, which are useful for searching the role of individual proteins in such a comlicated process of excitation energy transfer. The intensity of irradiance and temperature changes have a great impact on structure of complexes. The last chapter of theoretical part is dealing with influence of mentioned stress factors on PPC, but also on thylakoid membranes and leaves. In experimental part of this work, the low temperature (77K) emission and excitation spectra of the barley leaves (Hordeum vulgare L.) and its mutant chlorina f2 addapted to high and low irradiance were measured. Besides intact material the spectra of thylakoid membrane suspension and pigment-protein complexes separated by PAGE analysis were measured. The content of pigments in thylakoid membrane suspension and in electroforetically separated pigment-protein complexes was determined using HPLC methods and absorption spectroscopy. Further, the influence of different degrees of hight temperature and irradiance to efficiency of excitation energy transfer was studied on Norway spruce (Picea abies (L.) Karst.) growing in the growth chamber. Low and room temperature (293K) fluorescence spectra were measured on detached needles, together with pigment analysis from suspensions of needles perforged by HPLC.
