Abstract:

V interfázním jádře jsou chromosomy uspořádány do samostatných oblastí: chromosomálních teritorií. U těchto teritorií je možné pozorovat jisté radiální uspořádání v závislosti na genové hustotě. Některé chromosomy se také navzájem nacházejí v těsné blízkosti častěji než jiné, což může mít vliv na vznik translokací spojovaných s rakovinami. Tyto translokace mohou vést až ke změnám v uspořádání chromosomálních teritorií. Laminy typů A a B jsou hlavními složkami jaderné laminy, vláknité struktury nacházející se pod vnitřní jadernou membránou. Společně s mnoha dalšími proteiny se významně podílejí na organizaci chromatinu a mnoha jaderných procesech, včetně umlčování genů. Mutace v genech těchto proteinů způsobují závažná onemocnění včetně Hutchinsonova-Gilfordova syndromu předčasného stárnutí. Toto onemocnění je mimo jiné spojeno i se změnami v chromatinu.

Abstract:

Chromosomes are arranged in separate regions: chromosome territories in the interphase nucleus. Radial arrangement depending on the gene density can be observed in these territories. Some chromosomes are also located closer to each other than other chromosomes which may affect the formation of translocations associated with cancer. These translocations can lead to changes in the organization of chromosomal territories. A and B types lamins are the major components of the nuclear lamina, the filamentous structure located under the inner nuclear membrane. Together with many other proteins, they significantly partake of the organization of chromatin and many nuclear processes including gene silencing. Mutations in the genes of these proteins cause serious illnesses including Hutchinson-Gilford Progeria Syndrome. This disease is also associated with chromatin changes.
