Application of biosensors for detection of cholinesterase inhibitors

Abstract:

Tato diplomová práce se zabývala testováním inhibitorů cholinesteras pomocí biosensorů. Michaelisova konstanta, mezná rychlost a IC50 byly stanoveny spektrofotometrickou Ellmanovo metodou a elektrochemickou metodou (square-wave voltametrie). Získali jsme rozdílné výsledky v důsledku odlišných reakčních podmínek a jiné detekce. Dále jsme hledali sensor, který bude poskytovat nejlepší odezvu po imobilizaci enzymu metodou adsorpce a zesíťování glutaraldehydem. Následně byl pro testování inhibitorů acetylcholinesterasy vybrán nejlepší sensor, který se vyznačoval stabilními výsledky a odezvou, která neklesala v závislosti na čase. Tyto vlastnosti měl Pt-AgPd/Pt sensor. Naopak sensory s uhlíkovou měrnou elektrodou byly nevhodné, protoţe se na ně nepodařilo imobilizovat enzym. Po analýze jednotlivých inhibitorů acetylcholinesterasy byla stanovena jejich inhibiční účinnost a typ inhibice. Jako inhibitor s nejvyšší inhibiční účinností byl identifikován inhibitor 4. Abstract:

This diploma work deals with testing of some inhibitors of a cholinesterase using biosensors. Michaelis constant, marginal rate of enzymes and IC50 was determined with Ellman method and electochemical method (square-wave voltametry). Different results were obtained for both methods. This si probably caused by different reaction conditions and detection method. Then we searched a sensor with the best response after imobilization of enzyme with adsorption and cross-linking with glutaraldehyde. Consequently we choosed best sensor for testing inhibitors of acetylcholineserase. This sensor had stable results and response with no decreasing trend in the time. This quality had Pt-AgPd/Pt sensor. Conversely sensors with carbon working electrode weren´t suitable, because immobilization on this sensor wasn´t successfull. The effectivity of inhibitors and type of inhibition were determined. As a inhibitor with the highest efficiency of inhibition was identified inhibitor number 4.
