In vitro experiments and their relevance to situation in vivo - new trends in cell culture experiments

Abstract:

Tato práce srovnává tradiční způsoby testování toxicity s klasickým i moderním přístupem v testování in vitro a popisuje nejnovější trendy v experimentech in vitro. Jejím účelem bylo především objasnit a zdůvodnit možnou relevanci nových in vitro metod s testováním in vivo, její velkou důležitost a vyhlídky do budoucna. Obsahuje teoretickou a experimentální část. Mezi hlavní cíle teoretické části této bakalářské práce patří zmapování dostupné literatury týkající se dané problematiky, hlubší popis a vysvětlení nových trendů v experimentech s tkáňovými kulturami (výhody, nevýhody, možnost praktického využití) a jejich srovnání se situací in vivo. Metody a materiály kultivace buněčných kultur in vitro, upravování podmínek kultivace, snaha o vytváření 3D podmínek náhradou extracelulární matrix nebo použití podpůrných struktur jsou předměty teoretické části práce. Cílem experimentální části této bakalářské práce je prověřit jeden z alternativních in vitro modelů, tedy ko - kultivaci více typů buněk s využitím insertu a nejmodernějšího systému xCELLigence. Experiment má sloužit k přiblížení se k tématu a důkazu toho, že nejnovější metody nejsou pouze předmětem zahraničních autorů, ale i prakticky uskutečnitelné a funkční.

Abstract:

This academic paper compares conventional techniques of toxicity testing with classic and modern approach to testing in vitro. Also it describes the latest trends of experiments in vitro. The purpose of this paper was to point out the importance and the prospect for the future of new in vitro methods and also to clarify and to rationalize potential relevance of these methods with testing in vivo. This paper is divided into theoretical part and the experimental part. The main goal of theoretical part is reviewing of available literature related to the given issue, deeper description and explanation of new trends in experimenting with tissue cultures (advantages, disadvantages and the possibility of practical application) and its comparison with in vivo situation. Other issues in theoretical part are methods and materials for cultivation of cellular cultures in vitro, adjusting conditions of cultivation, effort to create 3D conditions by substitution of extracellular matrix or application of supportive structures. The main goal of the experimental part is to examine one of alternative in vitro models, the co-cultivation of varied types of cells with use of the latest system xCELLigence. The experiment should give the evidence that the latest methods are not just an issue for foreign authors, but it is a real and possible path.
