Abstract:

Diplomová práce se zabývá zavedením a optimalizací ELISA metody na magnetických částicích pro průkaz specifických protilátek proti biomarkerům Alzheimerovy choroby (AD), konkrétně protilátek proti beta amyloidu (A beta). Jelikož je při této metodě jako pevná fáze využíván povrch magnetických částic, byl nejprve optimalizován postup přípravy magnetického nosiče (tj. imobilizace antigenu) včetně jeho následné charakterizace. Dále byl optimalizován postup samotné ELISA metody na magnetických částicích, a to nejprve na modelovém systému s alfa-chymotrypsinem a následně na systému s A beta (1-42) jako vázaným antigenem. Řešena byla především otázka minimalizace nespecifické sorbce na povrch částic. Nakonec byl zavedený a zoptimalizovaný postup použit pro stanovení protilátek proti A beta v sérech pacientů s AD a zdravých kontrol.

Abstract:

The thesis deals with the implementation and optimalization of magnetic beads-ELISA method for detection of specific antibodies against biomarkers of Alzheimer's disease (AD), specifically of antibodies against beta amyloid (A beta). Since during this method the surface of magnetic beads is used as the solid phase, suitable procedure for the preparation of magnetic carriers (i.e. immobilization of antigen) including its subsequent characterization had to be optimized at the very beginning. Furthermore, the procedures of magnetic beads-ELISA themselves were optimized: first, on the model system with the ?-chymotrypsin and then on the system with A? (1-42) as a bound ligand. Finally, the antibodies against A? from the sera of AD patiens and healthy controls were measured by the optimazed and established method.
