Abstract:
V této práci jsem se pokusil separovat nukleové báze odvozené od cytosinu pomocí vysokoúčinné kapalinové chromatografie, tj. 5-methylcytosin a 5-hydroxymethylcytosin. Plánoval jsem použití monolitické kolony, z důvodu snahy o urychlení analýzy. Optimalizoval jsem tedy především složení mobilní fáze a to obsah methanolu, koncentraci pufru a hodnotu pH. Abstract:
The target of this thesis was to try to separate the nucleic bases derived from cytosine by using of high performance liquid chromatography, ie.derivatives were 5-methylcytosine and 5-hydroxymethylcytosin. I wanted to use monolithic columns, so the analysis should take a short time.
