Characterisation of antibodies against Bacillus subtilis using piezoelectric biosensor

Abstract:

V teoretické části byly popsány vlastnosti, protilátek, baktriálních analytů, objev Sauerbreyovy rovnice, vliv viskozity kapaliny na měření, funkce a zapojení piezoelektrických QCM systémů v průtkovém uspořádání. Dále byla náplní mé činnosti příprava QCM krystalu pro monitorování připravených vzorků Bacillus atrophaeus. Nejprve byla na zlaté elektrody křemenných výbrusů vytvořena thiolová monovrstva za pomoci cysteaminu. Dále byly elektrody aktivovány roztokem glutaraldehydu, který přispěl svou volnou aminovou skupinou k možnosti vazby protilátky. Byly celkem vytvořeny dva typy imobilizovaných krystalů – první typ měl kovalentně navázanou protilátku proti G+ bakeriím, typ druhý měl jako rekogniční vrstvu protilátku proti Bacillus subtilis. Měřeným analytem byly spory bakterie Bacillus atrophaeus v různých koncentračních ředěních. Následně byla prováděna regenerace biosenzoru za účasti roztoku glycinu (20mM, pH = 2). Doba celého měření byla v rozmezí 1-1,5hodiny, detekční čas byl několikrát prodlužován, abychom mohli porovnat jednotlivé grafické vyhodnocení, zda se určení změny frekvence rapidně nemění na době měření.

Abstract:

In the theoretical part, we describe properties of antibodies, bacterial analytes, discovery of the Sauerbrey´s equations, the influence of fluid viscosity on piezoelectric measurements, functions and involvement of quartz crystal microbalance (QCM) systems in the flow-through arrangement. Furthermore, the target of my training activities was preparation of QCM crystal samples for monitoring of Bacillus atrophaeus. First, thiol monolayer was created on the gold electrodes of quartz cuts by using cysteamine. Furthermore, electrodes were activated by glutaraldehyde solution, which transferred the free amino group to reactive aldehyde moieties suitable for covalent binding of antibodies. In this way, two types of immobilized crystals were obtained - the first type was modified with antibody against G+ bacteria, the other type had a recognition layer of antibody against Bacillus subtilis. The analyte was spore formed from Bacillus atrophaeus in different concentration dilutions. Subsequently, regeneration of the biosensor was achieved using glycine solution (20 mM, pH = 2). Measuring time was in the range of 1-1.5 hours, the incubation time of the sensor with microbes wa varried in order to observe and compare its effect on the measured change of frequency.
