Analysis of selected polyphenolic compounds in elderberry

Abstract:

První část diplomové práce byla zaměřena na optimalizaci chromatografické separace anthokyaninů v plodech černého bezu s využitím izokratické a gradientové eluce. Jako mobilní fáze byla použita směs metanol a voda s přídavkem kyseliny mravenčí. Optimalizovaná chromatografická separace byla ve spojení s UV detekcí využita k identifikaci a kvantifikaci anthokyaninů v extraktech z plodů černého bezu. Dále byl u extraktů stanoven celkový obsah anthokyaninů, celkový obsah polyfenolických látek a antioxidační kapacita pomocí různých spektrofotometrických metod. Výsledky získané těmito metodami byly statisticky porovnány s obsahem anthokyaninů stanovených pomocí kapalinové chromatografie. Na základě analýzy hlavních komponent a analýzy rozptylu byl zjištěn významný vliv úpravy vzorků po sběru (mražení/sušení). Největší korelace výsledků byla pozorována mezi pH diferenciální metodou a chromatografickým stanovením.

Abstract:

First part of this thesis was focused on optimization of chromatographic separation of anthocyanins in the fruit of elderberry using isocratic and gradient elution. The mixture of metanol and water with addition of formic acid was used as a mobile phase. Optimized chromatographic separation coupled with UV detection was used for identification and quantification of anthocyanins in extracts prepared from fruit of elderberry. Total anthocyanins, total polyphenolics and antioxidant capacity were determined using different spectrophotometric methods in all extracts. Results obtained by these methods were statistically compared with total anthocyanins determinated by liquid chromatography. Significant effect of treatment samples after collection (freezing/drying) was evaluated using principal component analysis and analysis of variance. The best correlation of results was observed between pH differential method and chromatographic determination.
