Determination of lipids in HDL2 and HDL3 fractions of EDTA plasma of diabetical patients

Abstract:

Předložená diplomová práce pojednává nejprve o lipidech, jejich metabolismu a propojení s metabolickými onemocněními jako diabetes mellitus, metabolický syndrom nebo onemocnění jater. Experimentální část se zabývá možnostmi optimalizace detekce při stanovení koncentrací jednotlivých podtříd lipidů pomocí tenkovrstvé chromatografie. Dále je zde popsáno využití metodiky plynové chromatografie k určení obsahu mastných kyselin a lipoidních enzymů ve frakcích HDL2 a HDL3 krevní plazmy diabetiků. Ve třetí výsledkové části jsou data statisticky zpracována a následně jsou diskutovány nalezené vztahy mezi zastoupením jednotlivých mastných kyselin a enzymů ?9, ?5, ?6 desaturáz a elongázy vůči závažnosti postižení diabetických pacientů. Metodou tenkovrstvé chromatografie se optimalizace detekce stanovení nepodařila a tato metoda bude využívána jen pro rozdělení lipidů do podtříd. Metodou plynové chromatografie nebyly zjištěny žádné signifikantní změny mezi frakcemi HDL2 a HDL3. Naopak byla potvrzena protektivní role palmitoleátu a vysvětlen byl pokles HDL?cholesterolu u diabetiků. Vztah ?6 desaturázy k onemocnění se také ukázal jako významný. Ze zjištěných korelací vyplývají možné protektivní vlastnosti tohoto enzymu.

Abstract:

The presented thesis deals with lipid metabolism and its link with metabolic diseases like diabetes mellitus, metabolic syndrome, or fatty liver disease at first. The experimental section discusses the possibilities of optimizing the detection in the determination of individual lipid subclasses concentrations by TLC. Furthermore, the methodology which is described here uses gas chromatography to determine fatty acids and enzymes in HDL2 and HDL3 fraction of EDTA plasma of diabetic patients. In the third part of results the data are statistically processed, and then there is a discussion about the relationships found between the proportion of fatty acids, enzymes ?9, ?5, ?6 desaturase and elongase and the severity of diabetes. The optimizing of detection in the determination of lipid concentration by TLC failed and this method is recommended only for separation into the lipids subclasses. Using gas chromatography there are found no significant differences between fractions HDL2 and HDL3. On the contrary the protective role of palmitoleate is confirmed and the decline of HDL ? cholesterol is explained. The relationship between ?6 desaturase and the disease has also emerged as significant. The observed correlations can indicate protective properties of this enzyme.
