Preparation, Characterization and 3D Printing of Magnetic Composite Systems Based on Thermoplastic Elastomers

Abstract:

Diplomová práce se zabývá chováním termoplastického elastomeru na bázi blokového kopolymeru poly(styrene-b-poly(ethylene-stat-butylene)-b-poly(styrene) (SEBS) plněného magnetickými částicemi karbonylového železa (CI) ve specifických poměrech (5-60 hm. %). Tyto kompozity mají vynikající schopnost chytrých materiálů reagovat na vnější podnět (magnetické pole) a také je lze připravovat ve formě scafoldů poskytujících vhodné prostředí pro kultivaci buněk. Teoretická část této práce se věnuje metodám 3D tisku a jeho využití pro přípravu scafoldů. Dále jsou zde popsány elastomerní systémy a materiály schopné odezvy na vnější podnět. Praktická část popisuje přípravu kompozitních materiálů obsahujících SEBS a CI částice a jejich následné charakterizování pomocí diferenciální snímací kalorimetrie, dynamické mechanické analýzy, rotační reometrie v oscilačním režimu bez a v přítomnosti magnetického pole. Popsány jsou dále procesy přípravy 3D tištěných scafoldů. V závěru práce je vyhodnocena cytotoxicita na základě extraktů a přímého kontaktu s buňkami, která zcela potvrzuje možnou aplikaci scafoldů jako substrátu v buněčném prostředí.

Abstract:

This master thesis deals with behaviour of the thermoplastic elastomer based on block copolymer poly(styrene-b-poly(ethylene-ran-butylene)-b-poly(styrene) (SEBS) filled with magnetic particles carbonyl iron (CI) at various filler content (5-60 wt. %). These composites have superior capability of smart systems responding on the external stimulus (magnetic field) and also can be fabricated in the form of scaffolds providing excellent substrate for cell culturing. Theoretical part of this thesis is dedicated to methods of 3D printing and its possibilities of the scaffold preparation. Furthermore, there is specific description of various elastomeric systems and stimuli responsive materials. The practical part is focused on the preparation of masterbatches containing SEBS and CI particles and their subsequent characterization using differential scanning calorimetry, dynamic mechanical analysis, rotation rheometry in oscillatory mode in absence as well as in the presence of magnetic field. The fabrication procedure of 3D printed scaffolds is carefully investigated. Finally, the evaluation of the scaffolds cytotoxicity, based on extracts and direct contact with cells clearly confirming their possible application as a substrate in the cells environment.
