A HPLC method for therapeutic drug monitoring of antipsychotics

Abstract:

Základem této bakalářské práce je literární rešerše na téma terapeutické monitorování léčby pomocí antipsychotik se zaměřením na klozapin. Teoretická část práce se zaměřuje na popis terapeutického monitorování plazmatických koncentrací léčiv v organismu obecně a dále cíleně na antipsychotika. Nedílnou součástí je pak popis základních fyzikálně-chemických vlastností klozapinu a jeho farmakodynamického a farmakokinetického chování v organismu. V práci je také uveden souhrn informací z publikací zabývajících se stanovením a monitorováním plazmatických koncentrací klozapinu a jeho metabolitů metodou vysokoúčinné kapalinové chromatografie s UV detekcí (HPLC-UV). Experimentální část vychází z poznatků vědeckých publikací, z nichž byly odvozeny počáteční podmínky HPLC-UV separace před samotnou optimalizací. Parametry metody byly optimalizovány na základě experimentálního testování a sledování vlivu změn na separované analyty, klozapin a jeho metabolity. U zvolené metody byla provedena krátká validace na základě vybraných parametrů a byla otestována její aplikace při separaci analytů obsažených v matricovém vzorku.

Abstract:

This bachelor's thesis is a literary research into therapeutic drug monitoring of therapy with antipsychotics with a specific focus on clozapine. The theoretical part of the thesis concentrates on describing therapeutic monitoring of plasma concentrations of medications in the organism generally and then more specifically of antipsychotics. A crucial part of this is a description of the basic physico-chemical properties of clozapine and its pharmacodynamic and pharmacokinetic behaviour in the organism. The thesis includes a summary of information from publications concerned with the assessment and monitoring of plasma concentrations of clozapine and its metabolites using the method High performance liquid chromatography with ultraviolet detection (HPLC-UV). The experimental part was produced using findings from scientific publications, from which the starting conditions of HPLC-UV separation were derived before optimization. The parameters of the method were optimized on the basis of experimental testing and observation of the effects of changes on the separated analytes, clozapine and its metabolites. For the chosen method, a short validation based on selected parameters was completed and its application tested during the separation of analytes contained in the matrix pattern.
