Synthesis and supramolecular properties of 1,8,13,20-tetraazaeicosane.

Abstract:

Cílem této práce bylo připravit dosud nepopsaný ligand 1,8,13,20-tetraazaikosan, jehož struktura vykazuje jistou analogií se sperminem, který s cucurbit[6]urilem tvoří vysoce afinitní komplexy s hodnotou Ka až 10^10 M^1. Celkem byly vyzkoušeny dva syntetické postupy. V prvním vystupoval jako klíčový intermediát diamid kyseliny jantarové, který se nám ovšem nepodařilo zredukovat na požadovaný amin. Během druhé syntetické strategie, zahrnující jako klíčový krok reakci 1,4-dibrombutanu s monochráněným hexamethylendiaminem, se objevily další překážky, které představovaly problémy se separaci produktu od výchozí látky. Po nalezení účinné separační metody byl izolován a charakterizován čistý produkt, jehož struktura ovšem neodpovídala titulnímu ligandu. Pro objasnění strukturní povahy připravené sloučeniny, byly provedeny supramolekulární experimenty pomocí NMR, ITC a ESI-MS. V budoucnu bude nezbytné modifikovat podmínky SN reakce, a to například úpravou poměru reakčních komponent, zvýšením teploty či prodloužením reakční doby.

Abstract:

This work aimed to prepare a previously undescribed ligand, 1,8,13,20-tetraazaeicosane, whose structure shows some analogy to spermine, which forms high-affinity complexes with Ka cucurbit[6]uril with values up to 10^10 M^1. Two alternative synthetic procedures leading to the title ligand were tested. A diamide of succinic acid was a key intermediate in the first approach. Unfortunately, we were not able to reduce it to the desired diamine. During the second synthetic strategy, additional obstacles were encountered that presented problems in the efficient separation of the product from the starting material. After finding an efficient separation method, a pure product was isolated whose structure didn´t match that of the title ligand. To elucidate nature of the prepared compound, supramolecular experiments were performed using NMR, ITC and ESI-MS. In the future, it will be necessary to modify the conditions of the SN reaction, for example, by adjusting the ratio of the reaction components, increasing the temperature or increasing the reaction time.
