Molecular diversity in honey bees evaluated by microsatellite loci

Abstract:

Předmětem této diplomové práce bylo studium diverzity molekulárních markerů u včely medonosné (Apis mellifera). Genetická diverzita byla hodnocena pomocí čtyř mikrosatelitních markerů z intergenových oblastí u celkem 114 vzorků klasifikovaných do devíti populací dle geografického původu. Cílem byla především optimalizace molekulárně genetických laboratorních metod, a následné statistické vyhodnocení získaných výsledků. V teoretické části práce je včela medonosná popsána z hlediska původu, poddruhů, morfologie, molekulární i populační genetiky. Všechny tyto znalosti byly následně uplatněny v experimentální části, která pojednává o optimalizaci molekulárních analýz, bioinformatickém zpracování genetických dat, a diskutuje problematiku, která v průběhu analýz může nastat. V rámci testovaného souboru bylo detekováno 38 různých alel. Jako nejvíce polymorfní se prokázal marker A107 a UN351. Výsledky na úrovni jednotlivých vzorků ukazují na vysoký stupeň variability, naproti tomu mezi populacemi příliš vysoká diverzita zaznamenána nebyla.

Abstract:

The aim of this study was the evaluation of molecular markers diversity in the honey bee (Apis mellifera). Genetic diversity was assessed using four microsatellite markers from intergenic regions in 114 samples classified to nine populations according to geographical origin. The main point was to optimaze molecular laboratory methods and then statistical evaluation of the obtained results. In the theoretical part of this work was described origin, subspecies, morphology, molecular and population genetics of honey bee. All this knowledge was applied in experimental part, which is about optimization of molecular analysis, bioinformatics processing of genetic data, and discusses the issues that may happen during the analysis. In the tested set of samples were detected 38 different alleles. Markers A107 and UN351 proved to be he most polymorphic. The results at level of individuals show a high degree of variability, on the other hand, between populations was not too high diversity recorded.
