Use of HLA antigens as predictive biomarkers of pharmacological response

Abstract:

Hlavní histokompatibilitní systém (MHC) u člověka je odpovědný nejen za správné fungování imunitního systému, ale také za kompatibilitu transplantovaných tkání, či reakci organismu na léčiva. V úvodu práce je proto stručně popsaná charakteristika HLA systému, jeho funkce v organismu a klasifikace antigenů dle jednotlivých tříd. Další část je věnována genetice a genomice (obor genetiky zabývající se studiem genomů), jejich úloze při predikci farmakologické odpovědi a výskytu nežádoucích účinků. V poslední části práce jsou popsány asociace mezi konkrétními HLA molekulami a nežádoucími účinky indikovanými léčivy.

Abstract:

The major human histocompatibility system (MHC) is responsible not only for the correct functioning of the immune system but also for the compatibility of the transplanted tissues or the response of the body to the drug. The introduction of the thesis is therefore a brief description of the HLA system its function in the organism and classification of antigens according to individual classes. The next part is devoted to genetics and genomics (study of genetics dealing with the study of genomes) their role in prediction of pharmacological response and occurrence of undesirable effects. The last part of the thesis describes associations between specific HLA molecules and adverse drug reactions.
