Analysis of circulating microRNAs in pancreatic cancer patients

Abstract:

Karcinom pankreatu patří mezi maligní onemocnění s nejvyšší mortalitou. Jedním z důvodů vysoké mortality tohoto onemocnění je pozdní diagnostika v pokročilém či dokonce metastatickém stádiu, které již neumožňuje radikální chirurgický zákrok. Z tohoto důvodu je potřeba hledat nové specifické biomarkery, které by umožnily spolehlivě diagnostikovat nádory pankreatu v časných stádiích. Jako vhodná varianta neinvazivních diagnostických biomarkerů pro nádorová onemocnění se jeví cirkulující mikroRNA přítomné i v krevní plazmě či séru. MikroRNA jsou krátké nekódující RNA, jejichž hlavní funkcí je post-transkripční regulace genové exprese. MikroRNA jsou zapojeny do mnoha fyziologických i patologických procesů včetně kancerogeneze a jejich význam v patogenezi karcinomu pankreatu byl opakovaně potvrzen. Tato bakalářská práce se věnuje analýze cirkulujících mikroRNA v plazmě pacientů s karcinomem pankreatu. Pomocí kvantitativní Real-Time PCR byla stanovena exprese vybraných mikroRNA (miR-17-5p, miR-21-5p, miR-126-5p, miR-155-5p, miR-375 a miR-1290) v plazmě pacientů s karcinomem slinivky a zdravých dárců. Jako nejvhodnější endogenní kontrola pro normalizaci dat byla identifikována miR-106a. Normalizovaná exprese všech studovaných mikroRNA byla signifikantně zvýšená u pacientů s karcinomem pankreatu a byl tedy potvrzen potenciál těchto cirkulujících mikroRNA sloužit jako diagnostické biomarkery odlišující pacienty s nádorem pankreatu od zdravých jedinců.

Abstract:

Pancreatic cancer is one of the malignancies with the highest mortality rate. One of the reasons for the high mortality rate of this disease is late diagnosis at an advanced or even metastatic stage when the radical surgical resection is not possible anymore. For this reason, it is necessary to search for new specific biomarkers that would enable a reliable diagnosis of pancreatic cancer at an early stage. Circulating microRNAs, which are present especially in blood plasma or serum, seem to be suitable non-invasive diagnostic biomarkers for cancer detection. MicroRNAs are short non coding RNAs, which function in post-transcriptional regulation of gene expression. MicroRNAs are involved in many physiological and pathological processes including carcinogenesis and their importance in the pathogenesis of pancreatic cancer has been repeatedly confirmed. This bachelor’s thesis deals with the analysis of circulating microRNAs in the plasma of pancreatic cancer patients. The expression of selected microRNAs (miR-17-5p, miR-21-5p, miR-126-5p, miR-155-5p, miR-375 and miR-1290) in the plasma of pancreatic cancer patients and healthy donors was determined using quantitative Real Time PCR. MiR-106a was identified as the most suitable endogenous control for data normalization. The normalized expression of all studied microRNAs was significantly elevated in pancreatic cancer patients and the potential of these circulating microRNAs to serve as diagnostic biomarkers distinguishing pancreatic cancer patients from healthy controls was thus confirmed.
