Imunitní odpověď prasečích makrofágů na in vitro infekce vyvolané invadujícími a neinvadujícími bakteriemi z čeledi Enterobacteriaceae

Immune response of porcine macrophages to in vitro infections induced by invasive and noninvasive bacteria from the Enterobacteriaceae family

Abstract:

Salmonella využívá při jejím intracelulárním parazitismu dva hlavní soubory genů, které nebyly nalezeny u Escherichia coli. Tyto geny jsou uspořádány do ostrovů patogenity 1 a 2 (tzv. SPI-1 a SPI-2) a podílí se na sestavování a funkci sekrečního systému typu 3 (T3SS), který je nutný pro invazi do hostitelských buněk a přežívání uvnitř. Tento poznatek souvisí s invazivností, Salmonella je fakultativně intracelulární invazivní baktérie, zatímco E. coli je většinou extracelulární neinvazivní baktérie. Hostitelská buňkami zprostředkovaná imunita (CMI), která je charakteristická interakcí makrofágů a Th1 lymfocytů, se výraznou měrou podílí na odstraňování intracelulárních patogenů jako je Salmonella. Makrofágy můžou zničit patogena a hrají zásadní roli v rozvoji, kontrole a odeznění salmonelózy v hostiteli. Avšak k dispozici je pouze omezené množství informací týkajících se role SPI-1 a SPI-2 při interakci salmonely s prasečími makrofágy. Proto jsme v naší studii nejprve infikovali permanentní prasečí makrofágovou buněčnou linii (3D4/31) divokým kmenem S. Typhimurium (WT) a jejími delečními mutanty ve SPI-1 a SPI-2 (konkrétně hilA a ssrA mutanti) a určili jsme jejich prozánětlivou odpověď pomocí kvantitativní RT-PCR v reálném čase (qRT-PCR). Tento experiment byl následně rozšířen o interakci 3D4/31 s enterotoxigenní E. coli (ETEC) chovající se jako deleční mutant ve SPI-1, který je neschopný invaze do hostitelských buněk. Nejprve bylo zjištěno, že WT a ssrA mutant indukovali signifikantně vyšší expresi mRNA pro IL-8, TNF-α a GM-CSF v 3D4/31 buněčné linii ve srovnání s hilA mutantem, ale pouze v přítomnosti prasečího séra. Dále jsme zjistili, že ETEC vyvolala produkci přibližně stejného množství IL-8 a TNF-α jako hilA mutant ve 3D4/31 buněčné linii. Vzhledem k tomu, že permanentní buněčné linie obvykle ztratí mnoho svých původních vlastností, v závěrečné studii jsme použili primární buňky reprezentované prasečími plicními alveolárními makrofágy (PAMs). Pro získání primárního celkového pohledu na odpověď prasečích makrofágů na infekci divokým kmenem byla použita technika microarray. Získané výsledky jsme použili v závěrečném experimentu, kdy jsme infikovali PAMs divokým kmenem a jeho delečními mutanty v celém souboru genů vymezující SPI-1 a SPI-2 (ve zkratce ∆SPI-1 a ∆SPI-2 mutanti) a určili jsme jejich imunitní odpověď pomocí qRT-PCR. V závěrečné sekci jsme zjistili, že PAMs odpovídaly na infekci ∆SPI-1 mutantem signifikantně vyšší expresí prozánětlivých (IL-1β, IL-8, TNF-α a GM-CSF) a Th17 cytokinů (IL-23) ve srovnání s WT a ∆SPI-2 mutantem. A naopak ∆SPI-1 mutant vyvolal signifikantně nižší expresi IL-4R, TIMP-1, TFRC a IL-7 v PAMs ve srovnání s WT a ∆SPI-2 mutantem. Celkově vzato, tato data zdůrazňují významnou roli genů kódovaných ze SPI-1 oblasti v časné fázi infekčního procesu, které se podílí nejenom na invazi, ale také na potlačení prozánětlivé a Th17 signální dráze a stimulaci makrofágů směrem k M2 polarizaci.

Abstract:

Salmonella harbours two major clusters of genes involved in its intracellular parasitism which have not been found in Escherichia coli. These genes are clustered to Salmonella pathogenicity islands 1 and 2 (SPI-1 and SPI-2) and are involved in the assembly and function of type III secretion system (T3SS) necessary for invasion into, and survival inside, the host cells, respectively. This knowledge is related to the invasiveness: Salmonella is a facultative intracellular invasive bacterium, contrary to E. coli, which is mostly an extracellular noninvasive bacterium. The clearance of intracellular pathogens such as Salmonella by a host is strongly dependent on cell-mediated immunity (CMI) which is characterized by the interactions between macrophages and Th1 lymphocytes. Macrophages can destroy the pathogen and play a crucial role in the development, control and clearance of salmonellosis from a host. However, only limited information is available on the role of SPI-1 and SPI-2 in the interaction of Salmonella with porcine macrophages. In our study, we therefore first infected permanent porcine macrophage cell line (3D4/31) with wild-type S. Typhimurium (WT) and its SPI-1 and SPI-2 deletion mutants (hilA and ssrA, respectively) and determined their proinflammatory response by quantitative real-time RT-PCR (qRT-PCR). This experiment was subsequently extended to the interaction of 3D4/31 with enterotoxigenic E. coli (ETEC) behaving like the deletion mutant in SPI-1 incapable of invasion into the host cells. First, it was found that WT and ssrA mutant induced significantly higher mRNA expression of IL-8, TNF-α and GM-CSF in 3D4/31 cell line when compared with the hilA mutant only in the presence of pig serum. Second, ETEC induced the production of similar amounts of IL-8 and TNF-α like hilA mutant in 3D4/31 cell line. Due to the fact that permanent cell lines usually have lost many of their original characteristics, we used primary cells represented by porcine pulmonary alveolar macrophages (PAMs). To get initial overview of PAMs response to the WT, we used the microarray analysis. The results obtained were applied to the last experiment where we infected PAMs with WT and its deletion mutants in whole SPI-1 (∆SPI-1) and SPI-2 (∆SPI-2) clusters of genes and determined their immune response by qRT-PCR. In the last section, we found that PAMs responded to the infection with ∆SPI-1 by a significantly higher expression of proinflammatory (IL-1β, IL-8, TNF-α and GM-CSF) and Th17 cytokines (IL-23) when compared with the WT or ∆SPI-2. Conversely, ∆SPI-1 mutant induced significantly lower expression of IL-4R, TIMP-1, TFRC and IL-7 in PAMs when compared with the WT or ∆SPI-2 mutant. Taken together, this data highlighted the important role of SPI1-encoded genes early in the infection process, not only in the invasion, but also in the suppression of proinflammatory and Th17 signaling and stimulation of M2 macrophage polarization.
