Disruption of thyroid hormone signaling by environmental pollutants

Abstract:

Thyroidní signální dráha a její narušování je v posledních letech před-mětem studia mnoha vědeckých publikací. Thyroidní hormony hrají důležitou roli v energetickém metabolismu, diferenciaci tkání, či termo-regulaci. Jejich význam roste zejména u vyvíjejících se plodů, které při-jímají thyroidní hormony přes placentu od matky a potřebují je zejména ke správnému vývoji nervové soustavy. Nedostatek hormonů pak může vést k závažným defektům v důsledku nesprávného vývoje mozku. Tento nedostatek může způsobovat široká škála thyroidních disruptorů v prostředí, kterým je organismus denně vystaven. Tato práce shrnuje poznatky o fungování thyroidní signální dráhy, její disrupci a běžně dostupných disruptorech. V praktické části je pak provedena optimalizace kultivace buněčné linie GH3, běžně využívané k in vitro testování účinků thyroidních disruptorů. Následně je na buněčné linii otestován účinek thyroidního hormonu trijodthyroninu metodou tzv. T-screenu, pro získání křivky dávka-odpověď a potvrzení funkčnosti testu. Na tento test je navázáno T-screenem, vystavujícím buňky léčivu difenylhydantoinu. Ve výsledcích T-screenu lze pozorovat inhibici růstu buněk, což potvrzuje thyroidně disruptivní vlastnosti tohoto léčiva. Bližší inhibiční vlastnosti této látky je však ještě nutné blíže prozkoumat.

Abstract:

Thyroid signalling system and its disruption is a subject of many scien-tific publications in last years. Thyroid hormones play an important role in energy metabolism, differentiation of tissues and thermoregulation. Their significance increases for fetus during pregnancy, because they accept thyroid hormones from mother, over the placenta and need it for their neurodevelopment. The deficiency of thyroid hormones can cause serious defects on mental health. The deficiency of the hormones can be the consequence of thyroid disruptors in the environment, where the organism is daily exposed. In this thesis we review the knowledge about function of thyroid signalling system, its disruption and available disruptors. In the experimental section we optimize the cultivation of GH3 cell line, which is usually used for testing of thyroid disruptors effects in vitro. Then, we test the effect of thyroid hormone triiodothyronine with T-screen method, for obtaining a dose-response curve and confirm the function on the method. In connection with this experiment, we continue with T-screen focused on effect of a drug diphenylhydantoin on the GH3 cells. The results of T-screen describe the inhibition of growth of the cells and confirm thyroid disruptive properties of the drug. A more detailed inhibition property of this substance needs to be a subject of next exploration.
