The role of selected adenylate cyclase isoforms in the regulation of lymphocyte functions

Abstract:

Adenylátcyklázy (AC) tvoří skupinu enzymů, které katalyzují tvorbu cyklického adenosinmonofosfátu (cAMP). Molekuly cAMP zastávají funkci sekundárního posla. cAMP signální dráha je důležitá pro spoustu buněk a procesů. V kontextu imunitního systému a T lymfocytů se dlouho uvažovalo o supresivní funkci cAMP. Momentálně se ví, že má i pozitivní vliv. Přechodně zvýšená hladina cAMP je nezbytná a podporuje aktivaci T pomocných (Th) lymfocytů, zatímco dlouhodobě zvýšená nebo snížená hladina cAMP má na vybrané subpopulace Th lymfocytů rozdílný vliv. Možná regulace hladiny cAMP přímou modulací na AC by mohla mít potenciální využití při léčbě autoimunitních a zánětlivých onemocnění. Cílem bakalářské práce bude shrnout aktuální dostupné informace, popsat cAMP signální dráhu se zaměřením na AC a vliv cAMP na aktivaci a diferenciaci Th lymfocytů, především na Th17 a T regulační (Treg) lymfocyty.

Abstract:

Adenylate cyclases (AC) are a group of enzymes that catalyse the synthesis of cyclic adenosine monophosphate (cAMP). cAMP molecules provide a secondary messenger function. The cAMP signalling pathway is important for many cells and processes. In the context of the immune system and T cells, the suppressive function of cAMP has been considered for a long time. It is now known, that cAMP has a positive effect as well. Transiently increased level of cAMP is necessary and promotes activation of T helper (Th) cells, while the long-term increased or decreased level of cAMP has different effect on selected Th subsets. Possible regulation of cAMP level through direct modulation of AC could have potential use in treatment of autoimmune and inflammatory diseases. The aim of this thesis will be to summarize the current available information, describe the cAMP signalling pathway with a focus on AC and the effect of cAMP on Th cells activation and differentiation, especially on Th17 and T regulatory (Treg) subsets.
