Biosensors and other methods for detection of inhibitors of cholinesterases

Abstract:

V tejto práci boli testované rôzne spôsoby imobilizácie cholinesteráz na povrch elektrochemických senzorov typu tlačených mikroelektród. Pripravené biosenzory boli použité pri testovaní koncentračného rozmedzia, časovej stability a následne na stanovenie inhibičnej účinnosti vybraných inhibítorov. Boli stanovené kinetické parametre a diskutoval sa vplyv imobilizácie na ich hodnotu. Na stanovenie inhibičnej účinnosti boli použité tri metódy s viacerými modifikáciami. Elektrochemická metóda (enzým prítomný v roztoku a enzým imobilizovaný na senzor), spektrofotometrická metóda (komerčné zakúpené enzýmy a enzýmy z ľudského zdroja) a pH-statová metóda s ľudskými enzýmami. Celkovo bolo testovaných 253 inhibítorov prevažne zo skupiny karbamátov a salicylanilidov. U všetkých látok bola stanovená inhibičná koncentrácia a rozdeľovací koeficient pomocou programu ChemSketch. Experimentálne bol stanovený rozdeľovací koeficient u 59 látok v systéme 1-oktanol a voda. Za účelom získania interakčných modelov s využitím molekulového modelovania bolo použitých 22 vybraných inhibítorov.

Abstract:

In this study different kinds of immobilization techniques onto the surface of electrochemical sensors, screen-printed electrodes, were tested. Prepared biosensors were used for testing of the concentration range, stability in time and subsequently for determination of inhibition efficiency of tested inhibitors. The kinetic parameters were determined and the effect of immobilization was discussed. For determination of inhibition efficiency three methods with modifications were used. Electrochemical method when enzyme was present in solution or immobilized onto the surface of sensor, spectrophotometric method with commercially bought enzymes or enzymes from human blood and pH-stat method with using of human enzymes were used. Totally 253 inhibitors from the group of carbamates and salicylanilides were tested. The inhibitory concentration and the partition coefficient with using of program ChemSketch were determined. The partition coefficient in system 1-octanol and water was determined for 59 inhibitors. In order to obtain the interactions models for selected 22 inhibitors the molecular modelling was used.
