Abstract:

Náplň této diplomové práce je součástí projektu zaměřeného na studium interakčních partnerů glyceraldehyd-3-fosfátdehydrogenázy (GAPDH) u bakterie Francisella tularensis s cílem odhalit nové neenzymatické funkce tohoto proteinu. V rámci této práce byly potvrzeny interakce enzymu GAPDH s bakteriálními proteiny vybranými na základě předchozí interakční analýzy. Experimentální část spočívala v zaklonování genů kódujících vybrané proteiny do expresních vektorů, následné přípravě a purifikaci rekombinantních proteinů a potvrzení interakcí metodou "Solid-phase ligand binding assay".

Abstract:

The objective of this diploma thesis is part of a project aimed at the study of interaction partners of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) in the bacterium Francisella tularensis in order to reveal new non-enzymatic functions of this protein. The interactions of the GAPDH enzyme with bacterial proteins selected on the basis of previous interaction analysis were confirmed in this thesis. The experimental part consisted of cloning genes encoding selected proteins into expression vectors, subsequent preparation and purification of recombinant proteins and confirmation of interactions using the "Solid-phase ligand binding assay" method.
