Analysis of anti-cancer properties of wedelolactone

Abstract:

Tato diplomová práce je zaměřena na poznání vlivu wedelolaktonu na linie prsních karcinomů. Wedelolakton je přírodní polyfenolická látka, která byla izolována z rostlin Wedelia chinensis a Eclipta prostata. Byly prokázány antiproliferační a proapoptotické účinky wedelolaktonu na několika typech nádorů in vitro a in vivo. Prsní karcinomy mohou být z hlediska exprese estrogenového receptoru (ER) rozděleny do dvou kategorií. ER-pozitivní nádory vykazují expresi ER, jejich růst je závislý na stimulaci estradiolem a jsou citlivé na estrogenové antagonisty. ER-negativní nádory nejsou závislé na stimulaci estradiolem a k terapii estrogenovými antagonisty jsou rezistentní. Vliv wedelolaktonu na prsní karcinomy jsme sledovali na ER-pozitivních liniích- MCF-7 a T47D a na ER-negativní linii MDA-MB-231. Zjistili jsme, že se wedelolakton v nízkých koncentracích (nM) chová jako estrogenový agonista a stimuluje růst ER-pozitivních nádorů. Tento stimulační efekt je ER-dependentní. Apoptóza indukovaná wedelolaktonem je ER-independentní a byla indukována vysokými koncentracemi (μM) u ER-pozitivních i negativních linií. Naše výsledky ukazují, že by wedelolakton mohl být využíván v terapii ER- negativních prsních karcinomů.

Abstract:

The aim of this Thesis is to clarify the influence of the wedelolactone on breast carcinoma cell lines. Wedelolactone is a natural polyphenolic compound, isolated from Wedelia chinensis and Eclipta prostata. Wedelolactone was found to suppress cellular growth and to induce apoptosis of several types of cancer cells in vitro and in vivo. Breast carcinomas can be divided into 2 groups according to estrogen receptor (ER) expression. ER-positive breast carcinomas express ER, their growth depends on estradiol stimulation a they are sensitive to estrogen antagonists. Growth of ER-negative breast carcinomas is not dependent on estradiol and they are resistant to therapy with estrogen antagonists. Effect of wedelolactone on breast cancer was analysed in ER-positive cell lines MCF-7 and T47D and ER-negative cell line MDA-MB-231. We found that in low concetration (nM) wedelolactone acts as estrogen agonist and stimulates growth of ER-positive cancer cells. This stimulating effect is ER-dependent. On the other hand, wedelolactone-induced apoptosis is ER-independent and was observed after treatment with high concentrations (μM) in both ER-positive and negative cancer cell lines. Our data suggests, that wedelolactone can be used in therapy of ER-negative breast carcinomas.
