Abstract:

Předkládaná disertační práce je souborem experimentálních výsledků vztahujících se primárně ke dvěma vybraným proteinům, které jsou zapojeny v cytokininovém metabolismu a signalizaci u rostlin. Prvním ze studovaných proteinů je enzym β-D-glukosidasa. V rámci výzkumu specificity tohoto enzymu byla publikována studie efektivity saturační mutageneze vybrané pozice W373. Součástí této publikace byla také purifikace mutantních forem enzymu, získaných jako rekombinantní proteiny cestou heterologní genové exprese v E. coli a určení počáteční rychlosti katalýzy vytvořených mutantů enzymu za použití umělého i přirozeného substrátu. β-D-glukosidasa se stala modelovým enzymem pro vytvoření automatizovaného postupu pro měření enzymové kinetiky, a to za použití jak přirozených, tak umělých substrátů. Měření enzymové kinetiky za použití umělého substrátu je založeno na kvantifikaci aglykonu vznikajícího při enzymatické reakci. Při měření kinetiky za použití přirozeného substrátu je kvantitativně stanoven druhý produkt enzymatické reakce, β-D-glukopyranosa. Druhým studovaným proteinem je cytokininový receptor CRE1/AHK4. Přirozenými agonisty tohoto receptoru jsou cytokininy, které v rostlinách vznikají mimo jiné hydrolýzou zásobních O-β-D-glukosidů za katalytického působení již zmíněné β-D-glukosidasy. Interakce receptoru CRE1/AHK4 s potencionálními agonisty/antagonisty byly studovány za pomocí arteficiálního systému E. coli KMI001. Modifikovaná bakteriální signální dráha, která je spouštěna transformovaným CRE1/AHK4 receptorem, končí expresí β-D-galaktosidasy. Aktivitu tohoto enzymu lze sledovat např. flourimetricky stanovením produktu enzymatické reakce. V rámci studia tohoto receptoru byla vyvinuta a publikována skríningová metoda sloužící k identifikaci jeho agonistů a antagonistů. Metoda rovněž zahrnuje detekci látek, které ruší stanovení fluorescenčního produktu β-D-galaktosidasové reakce. Celý experimentální postup byl rovněž automatizován a zpracován ve formě podané patentové přihlášky.

Abstract:

This dissertation is a set of experimental results related to two selected proteins that are involved in cytokinin metabolism and signaling in plants. The first protein is a β-D-glucosidase. During the determination of the specificity of this enzyme, an efficiency analysis of saturation mutagenesis in the pre-selected W373 position was published. This publication also included a protocol for the purification of mutant forms of the enzyme as recombinant proteins using heterologous gene expression in E. coli. Initial velocities of the created mutants were measured using artificial and natural substrates. β-D-glucosidase was developed as model enzyme to create an automated measurement procedure for enzyme kinetics using both natural and artificial substrates. Measurement of enzyme kinetics using an artificial substrate is based on the quantification of the aglycone formed during the enzymatic reaction. The second product of the enzymatic reaction, β D glucopyranose, was used to quantitatively determine kinetics while using a natural substrate. The second studied protein is the CRE1/AHK4 cytokinin receptor. The natural agonists of this receptor are cytokinins formed in planta, e.g., by hydrolysis of storage O-β-D-glucosides following the action of the aforementioned β-D-glucosidase. Interaction of the CRE1/AHK4 receptor with potential agonists/antagonists was studied using the artificial system set up in the E. coli strain KMI001. The system relies on a modified bacterial signaling pathway, which is triggered by the CRE1/AHK4 receptor, resulting in the expression of β-D-galactosidase. The activity of this enzyme can be monitored, for example, by fluorimetric determination of the enzymatic reaction product. Within the study of this receptor, a screening method to identify its agonists and antagonists was developed and published. The screening procedure also involved detection of compounds that interfere with the determination of the fluorescent product of the β-D-galactosidase reaction. The entire experimental procedure was also automated and formed the basis of a patent application.
