Deletion of prophages and redundant genes from the genome of <I>Pseudomonas aeruginosa</I> used for the production of lytic phages.

Abstract:

Pseudomonas aeruginosa je významným oportunním patogenem způsobujícím závažné nozokomiální infekce. Této bakterii je věnována stále větší pozornost, zejména v souvislosti s narůstající antibiotickou rezistencí. Alternativou léčby infekcí vyvolaných rezistentními bakteriemi je použití terapeutických fágů. Při aplikaci fágových preparátů je však nezbytné zajistit bezpečnost jejich podání pacientovi. K přípravě fágových preparátů je proto nutné mít dobře charakterizovaný bakteriální kmen, sloužící k pomnožování terapeutických fágů, a dále pak znát složení vzniklého fágového lyzátu. Teoretická část této práce je věnována mírným fágům, jejich schopnosti integrace do bakteriálního genomu za vzniku profága, důsledkům jimi způsobených infekcí, a možnostem eliminace nežádoucích genetických elementů v podobě profágových sekvencí. V rámci experimentální části byla na základě bioinformatické analýzy genomu bakterie Pseudomonas aeruginosa MB301, spolu s výsledky detekce a identifikace indukovaných profágů, provedena delece genu kódujícího fágovou integrázu.

Abstract:

Pseudomonas aeruginosa is an emerging opportunistic pathogen causing serious nosocomial infections. This bacterium is receiving increasing attention, especially in the context of rising antibiotic resistance. An alternative treatment for infections caused by resistant bacteria is the use of therapeutic bacteriophages. However, when administering bacteriophage preparations, it is essential to ensure the safety of their administration to the patient. For the development of phage preparations, it is therefore necessary to have a well-characterized bacterial strain used for the multiplication of therapeutic phages and to know the composition of the produced phage lysate. The theoretical part of this thesis is devoted to temperate phages, their ability to integrate into the bacterial genome and form prophages, the consequences of infections caused by them, and the possibility of eliminating unwanted genetic elements in the form of prophage sequences. In the experimental part, deletion of the gene encoding phage integrase was performed based on bioinformatic analysis of the genome of Pseudomonas aeruginosa MB301, along with the results of detection and identification of induced prophages.
