Physiological reactions of honey bee haemocytes under stress conditions.

Abstract:

The western honey bee Apis mellifera is an eusocial species of insect living in tightly interconnected societies. Structure of these societies enables easy transmission of pathogens. Therefore, bees employ a wide spectrum of immune system reactions for their defense. One of its main components is a cellular response performed by specialized immune cells – the haemocytes. It is known that during the infection, these cells participate in phagocytosis, nodulation, and encapsulation of the pathogens, although, little is known about potential changes they go through in response to infectious stress. One of the reasons for this is the lack of sufficiently reliable and effective methods for their analysis. This thesis utilizes flow cytometry to monitor physiological changes of honey bee haemocytes in response to two kinds of pathogens – an ectoparasite Varroa destructor and a bacterium Serratia marcescens. Both pathogens cause great changes in concentration, permeability, and viability of the haemocytes. While doing so, the variability of these changes is dependent on the season, which is caused by a distinct representation of short-living summer bees and long-living winter bees in beehives of our climate zone. In summer bees, dynamic change of the haemocytes has been clearly observed in response to bacterial infection, however, the winter bees did not show any observable change.

Abstract:

Včela medonosná, Apis mellifera, je eusociální druh hmyzu žijící v úzce propojených společenstvích, která svojí strukturou umožňují snadný přenos patogenů. Včely proto využívají široké spektrum reakcí imunitního systému na svoji obranu. Jednou z jeho hlavních složek je buněčná odpověď zajišťovaná specializovanými imunitními buňkami – hemocyty. Je známo, že tyto buňky se v průběhu infekce podílejí na fagocytóze, nodulaci nebo enkapsulaci patogenů, ale málo se ví o potenciálních změnách, ke kterým u nich v reakci na infekční stres dochází. Jedním z důvodů je nedostatek dostatečně spolehlivých a efektivních metod jejich analýzy. Tato práce proto využívá průtokovou cytometrii na sledování fyziologických změn hemocytů včel v reakci na dva druhy patogenů – ektoparazita Varroa destructor a bakterii Serratia marcescens. Oba patogeny v průběhu infekce způsobují významné změny v koncentraci, permeabilitě a viabilitě hemocytů. Variabilita těchto změn je přitom závislá na ročním období, což je způsobené odlišným zastoupením krátce žijících letních včel a dlouhověkých zimních včel ve včelstvech našeho podnebného pásu. Zatímco u letních včel byla v průběhu experimentů jasně pozorovaná dynamická reakce hemocytů na bakteriální infekci, zimní včely nevykazovaly žádnou pozorovatelnou změnu.
