Prolyl endoprotease from Aspergillus niger and its use in the analyse of proteins

Abstract:

V teoretické části této bakalářské práce se pojednává o proteolytických enzymech a jejich využití v proteomické analýze proteinů metodou hmotnostní spektrometrie. Dále je zde popsána rodina enzymů prolyloligopeptidas a prolylendoproteasa z Aspergillus niger. Zmíněny jsou zde zejména rozdíly mezi těmito enzymy a aplikace PSE v rozdílných odvětvích průmyslu. V experimentální části byla izolována prolylendoproteasa z Aspergillus niger s kyselým pH optimem z komerčního produktu Brewer?s Clarex za účelem vyhodnocení její možné aplikace v proteomice. Chromatografickou purifikací bylo dosaženo jediného proteinového pásu na gelu po SDS-PAGE (?sodium dodecyl sulfate polyacrylamide gel electrophoresis?) s molekulovou hmotností 63 kD odpovídající širokému maximu (m/z 58,061) v hmotnostním spektru MALDI (?matrix-assisted laser desorption/ionization?) TOF (?time-of-flight?) indikujícímu glykoprotein. Získaná data ukázala, že enzym je přítomen v jeho zpracované formě bez signálního peptidu a propeptidu. Spektrofotometricky bylo zjištěno pH optimum okolo 4,5 a byla pozorována vysoká teplotní stabilita. Štěpením proteinových standardů (12-200 kDa) pomocí PSE v roztoku byly získány peptidové směsi, které ukázaly preferenci štěpení za prolinovými a alaninovými zbytky a minoritní, ale významné štěpení za dalšími aminokyselinovými zbytky jako jsou např. G, L, R, S a Y. Štěpení včelího peptidu apidaecinu 1A obsahujícího 6 prolinových zbytků vedlo k detekci specifických peptidů končících prolinem, což bylo potvrzeno MALDI PSD (?post-source decay?) analýzou

Abstract:

The theoretical part of this bachelor thesis deals with proteolytic enzymes and their use in proteomic analysis of proteins by mass spectrometry. Furthermore the enzyme family of prolyl oligopeptidases is described together with prolyl endoprotease from Aspergillus niger. Major differences between these enzymes as well as the application of PSE in different branches of industry are mentioned. In the experimental part an acidic prolyl endoprotease from Aspergillus niger was isolated from a commercial product Brewer?s Clarex to evaluate its possible application in proteomics. The chromatographic purification yielded a single protein band in sodium dodecyl sulfate polyacrylamide gel electrophoresis providing an apparent molecular mass of 63 kDa and a broad peak (m/z = 58,061) in linear matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) mass spectrometry indicating a glycoprotein nature enzyme. The obtained data also show that the enzyme is present in its processed form, i.e. without putative signal and propeptide parts. A spectrophotometric activity assay method demonstrated a pH optimum around 4,5 and a high thermostability for the cleavage at the C-terminal part of proline residues. In-solution digestion of standard proteins (12-200 kDa) by PSE resulted in a number of peptides reflecting a major enzyme action on proline and alanine residues and a minor but significant action on some other residues like for example G, L, R, S and Y. The digestion of honeybee peptide apidaecin 1A comprising six proline residues led to the detection of specific peptides by proline as it was confirmed by MALDI post-source decay analysis
