Monitoring of resistance to antifungal agents at medically important yeasts

Abstract:

Nejčastějšími původci mykotických onemocnění jsou kvasinky rodu Candida. K jejich léčbě se používají antimykotika. Flukonazol patří k velmi často používaným antimykotikům, avšak díky jeho častému užívání došlo k rozvoji rezistence k tomuto léku. Cílem této práce bylo zjištění rozvoje rezistence k flukonazolu u kvasinek rodu Candida. Poté byly získané výsledky porovnány s literaturou. Hodnoceno bylo celkem 20 kmenů C. albicans, 20 kmenů C. glabrata, 20 kmenů C. tropicalis a 20 kmenů C. dubliniensis. Testováním indukce rezistence kmenů kvasinek k flukonazolu bylo zjištěno, že C. albicans rostla do koncentrace 25 µg/ml, C. glabrata do 50 µg/ml, C. tropicalis do 10 µg/ml a C. dubliniensis do 1 µg/ml. E-testem byla u kmenů původních i indukovaných stanovena minimální inhibiční koncentrace. Bylo prokázáno, že C. glabrata byla ze všech čtyř testovaných druhů nejvíce odolná vůči flukonazolu, naopak C. dubliniensis byla nejvíce citlivým druhem.

Abstract:

The most common causes fungal infections are a yeast of the genus Candida. To their treatment is used antifungal agents. Fluconazole is one of the most commonly used antifungal agents, but due to his frequent use have developed resistance to this drug. The aim of this study was to determine the development of resistance to fluconazole in Candida species. Then, the results were compared with the literature. Evaluated were a total 20 strains of C. albicans, 20 strains of C. glabrata, 20 strains of C. tropicalis and 20 strains of C. dubliniensis. Testing induction of resistance to fluconazole yeast strains was found that C. albicans grown to a maximum concentration 25 µg/ml, C. glabrata to 50 µg/ml, C. tropicalis to 10 µg/ml and C. dubliniensis to 1 µg/ml. By E-test was determined minimum inhibitory concentration for original and induced strains. It was shown that C. glabrata was from all four test species most resistant to fluconazole, while C. dubliniensis was the most sensitive species.
