Quantitative analysis of SMN1 gene

Abstract:

Spinální muskulární atrofie (SMA) je poměrně časté letalizující nervosvalové onemocnění s autozomálně recesivním typem dědičnosti. Incidence SMA je přibližně 1/5600. Onemocnění je způsobeno homozygotní delecí genu SMN1 (Survival of motor neuron 1). Cílem mé bakalářské práce bylo teoretické a praktické zvládnutí kvantitativní analýzy genu SMN1 a stanovení frekvence přenašečů SMA v Moravskoslezském kraji (MSK). S použitím dat Laboratoře Oddělení lékařské genetiky Fakultní nemocnice Ostrava jsem provedl retrospektivní studii pacientů vyšetřených v letech 2011 až 2016. U vybraného vzorku pacientů jsem provedl kvantitativní analýzu genu SMN1. Celkem bylo vyšetřeno 1389 pacientů. U 1267 byly nalezeny 2 kopie genu SMN (norma), u 83 pacientů 1 kopie genu SMN1 (přenašeči SMA) a u 11 pacientů byla detekována homozygotní delece genu SMN1 (pacienti s různým typem SMA). Pro určení počtu kopií genu SMN1 byly použity metody real-time PCR a MLPA (Multiplex ligation-dependent probe amplification). Frekvence přenašečů SMA v MSKbyla stanovena ze souboru pacientů, u kterých bylo vyšetření provedeno jako doplňkové. Frekvence přenašečů SMA je v MSK 3,099 %, což je v souladu s literárními údaji.

Abstract:

Spinal muscular atrophy (SMA) is quite common lethal neuromuscular disease with autosomal recessive type of heredity. Incidence of SMA is approximately 1/5600. This disease is caused by homozygous deletion of SMN1 (Survival of motor neuron 1) gene. The aim of my thesis was mastering theoretical and practial skills of quantitative analysis of SMN1 gene analysis and assessment of SMA carrier frequency in Moravian-Silesian region (MSR). I have conducted retrospective studies of patients examined over the years 2011 ? 2016 using the data of Department of Medical Genetics in University Hospital Ostrava. With a selected sample of patients I have conducted a quantitative analysis of SMN1 gene. A total of 1389 patients were examined. 1269 patients 2 copies of SMN gene were found (standard), with 83 patients 1 copy of SMN1 gene were found (SMA carriers) and with 11 patients homozygous deletion of SMN1 gene were detected (patients with diverse SMA subtype).For determination of SMN1 gene quantity real-time PCR and MLPA (Multiplex ligation-dependent probe amplification) methods were used. The SMA carriers frequency in MSR was determined from a group of patients, who had been examined additionaly. The SMA carriers frequency in MSR is 3,099 %, which is consistent with literature data.
