The relationship between candidate gene polymorphisms and mild cognitive impairment

Abstract:

BDNF (brain-derived neurotrophic factor) je protein, který sehrává důležitou úlo-hu v procesu neurogeneze. Snížená hladina BDNF je jednou z příčin patologických procesů neurodegenerativních onemocnění. Mírná kognitivní porucha je pokládá-na za předstupeň Alzheimerovy choroby. V rámci této práce bylo pomocí SNaPshot metody na kapilárním sekvenátoru analyzováno celkem 10 polymorfiz-mů genu pro BDNF u 127 pacientů s mírnou kognitivní poruchou. Analyzované polymorfismy genu pro BDNF byly zkoumány ve vztahu k výsledkům kognitivních testů a s různými onemocněními, které se u pacientů vyskytovaly, jako jsou hyper-tenze, ischemie, infarkt, jiné srdeční choroby, vysoká hladina cholesterolu, epilep-tické záchvaty, plicní onemocnění, zranění, alergie, depresivní porucha, jiná psy-chická porucha a nádorové onemocnění. Statisticky významné asociace byly nale-zeny s výsledky některých kognitivních testů. U sledovaných onemocnění byly na-lezeny asociace pouze mezi polymorfismy a vyšší hladinou cholesterolu a alergie-mi u pacientů s MCI. Výsledky této práce bude třeba ověřit na větším souboru osob.

Abstract:

BDNF (Brain Derived Neurotrophic Factor) is protein which plays an important role in whole procces of neurogenesis. The lower level of BDNF is one of the causes of phatologic processes in neurodegenerative diseases. Mild Cognitive Im-pairment is considered as prestage of Alzheimer’s disease. In this thesis 10 poly-morphisms of BDNF gene was analysed on capillary analyser using SNaPshot me-thod on group of 127 patients with Mild Cognitive Impairment. Analysed poly-morphisms of BDNF gene were examined in the relationship with results of cogni-tive test and various diseases such as hypertension, ischemia, myocardial infarcti-on, other heart diseases, high levels of cholesterol, epileptic seizures, lung disease, injuy, allergy, depression, other mental illness and tumor disease. Statistical asso-ciations were found with results of some cognitive tests. In examined diseases sta-tistictical associantions were found only between polymorphisms and higher le-vels of cholesterol and allergy in patiens with Mild Cognitive Impairment. The re-sults of this thesis need to be verify on larger group of patiens.
