Testing the efficicency of the selected recombinant lytic systems in bacterium Pseudomonas putida

Abstract:

Zavádění programovatelných lytických systémů do "buněčných továren" k dosažení časově regulovaného růstu buněk může být účinnou strategií pro vývoj udržitelných a levnějších biotechnologických procesů. Taková strategie by mohla zefektivnit nejen uvolňování intracelulárních produktů z buněk jak v tzv. downstream nebo upstream částech bioprocesu, ale také při konstrukci strukturně se měnících multikulturních populací. Teoretická část této diplomové práce je věnována především inženýrství a uplatnění programovatelných lytických systémů v moderních průmyslových biotechnologiích. V experimentální části byl zkonstruován nový rekombinantní lytický systém založený na bioinformaticky identifikovaném genu profágového původu. Dynamika nového lytického systému a dvou dalších systémů s odlišným mechanismem účinku byla ověřena v biotechnologicky atraktivním mikrobiálním hostiteli Pseudomonas putida za různých kultivačních podmínek. Účinnost tři lytických systémů byla porovnána a kriticky zhodnocena z hlediska funkčnosti a efektivity uvolňovaní reportérového proteinu z buňky do kultivačního média.

Abstract:

The introduction of programmable lytic systems into „cell factories“ to achieve temporal cell growth can be an effective strategy for developing sustainable, cheaper biotechnological processes. Such a strategy could streamline not only the release of intracellular substances from cells either in the downstream or upstream processing but also the construction of structurally changing multi-culture population. The theoretical part of this diploma thesis is mainly devoted to the engineering and application of programmable lytic systems in modern industrial biotechnology. In the experimental part, a novel recombinant lytic system based on a bioinformatically determined prophage gene was constructed. The lysis dynamics of the novel system and two other systems with different modes of action were screened in a biotechnologically attractive microbial host, Pseudomonas putida, under various culture conditions. The performance of the three lytic systems was compared and critically evaluated in terms of functionality and efficiency in releasing a reporter protein from the cell interior into the culture medium.
